Renin is a key enzyme in the renin-angiotensin system (RAS), a pathway which plays an important physiological role in blood pressure and electrolyte homeostasis. The origin of the RAS is believed to have accompanied early evolution of vertebrates. However, renin genes have so far only been unequivocally identified in mammals. Whether or not a bona fide renin gene exists in nonmammalian vertebrates has been an intriguing question of physiological and evolutionary interest. Using a genomic analytical approach, we identified renin genes in two nonmammalian vertebrates, zebrafish (Danio rerio) and pufferfish (Takifugu rubripes). Phylogenetic analysis demonstrates that the predicted fish renins cluster together with mammalian renins to form a distinct subclass of vertebrate aspartyl proteases. RT-PCR results confirm generation of the predicted zebrafish mRNA and its expression in association with the opisthonephric kidney of adult zebrafish. Comparative in situ hybridization analysis of wild-type and developmental mutants indicates that renin expression is first detected bilaterally in cells of the interrenal primordia at 24 h postfertilization, which subsequently migrate to lie adjacent to, but distinct from, the glomerulus of the developing pronephric kidney. Our report provides the first molecular evidence for the existence of renin genes in lower vertebrates. The observation that the earliest renin-expressing cells, arising during ontogeny of this teleost vertebrate, are of adrenocortical lineage raises an interesting hypothesis as regards the origin of renin-expressing cells in the metanephric kidney of higher vertebrates.

The toadfish,Opsanus tau, lacks renal glomeruli and macula densa, but has high renal renin activity and abundant granulated cells in renal arteries and arterioles. Reduction of blood pressure (BP) or blood volume by hemorrhage or vasodilatory drugs causes renin release, indicating that an intrarenal or extrarenal pressure- or volume-sensitive mechanism exists for controlling renin release in the toadfish. Thus, we examined whether 1) β-adrenergic receptor-mediated activation of renin release, and 2) calcium influx which may underlie the baroreceptor mechanism are involved in the cellular control of renin release. Acute injection of isoproterenol (1 μg/kg, n = 6) decreased BP and increased plasma renin activity (PRA) 4-5 fold in unanesthetized toadfish. Propranolol abolished both effects, but did not decrease basal PRA levels.In vitro superfusion of renal slices with bicarbonate Ringer's solution showed a steady secretion of renin, and addition of 50 mM K(+) (K(+) methylsulfate replacing NaCl, n = 10) to the superfusate markedly suppressed renin secretion. Nifedipine (10(-5) M, n = 8) completely restored the high K(+)-induced inhibition of renin secretion from renal slices, whereas isoproterenol (10(-4) M, n = 6) neither increased basal renin secretion nor restored K(+)-induced renin suppression. These results suggest that calcium influx may mediate inhibitory messages for renin secretion, while the β-adrenoceptor-mediated activation of granulated cells appears absent in toadfish.

Radioimmunoassay and characterization of renin‐angiotensin system in the fresh‐water turtle

We developed a radioimmunoassay (RIA) for plasma renin concentration (PRC) and characterized the renin-angiotensin system BAS) in the fresh-water turtle (A mydajaponica).Among three antisera to human angiotensin I (ANG I) had different specificities to ANG I analogues; the antiserum with the highest specificity was used to measure the PRC. The amount of ANG I generated from incubation of turtle renal extracts with turtle plasma was linear with respect to volume of renal extract and incubation time. Turtle renin cleaved heterologous substrates from nephrectomized rats and rabbits but had a higher affinity for renin substrate from rabbits. The generation of ANG I with heterologous substrate was temperature dependent (optimum-37°C) and the pH optimum was at 5.0-5.5. Furosemide (5 mgkg) did not affect PRC. Hemorrhage (1% and 3% of body weight) decreased blood pressure. The PRC did not change with the lower hemorrhage, but increased significantly with massive hemorrhage. Angiotensin I1 increased blood pressure with no effect in PRC. The PRC of fresh-water turtles transferred to 4 "C increased significantly.The RAS in the fresh-water turtle seems to be involved in the adaptation of environmental temperature rather than in the acute control of blood pressure although the mechanism is not known.
