Angiotensin-converting enzyme 2 inhibits high-mobility group box 1 and attenuates cardiac dysfunction post-myocardial ischemia

Abstract: High-mobility group box 1 (HMGB1) triggers and amplifies inflammation cascade following ischemic injury, and its elevated levels are associated with adverse clinical outcomes in patients with myocardial infarction (MI). Angiotensin-converting enzyme 2 (ACE2), a key member of vasoprotective axis of the renin-angiotensin system (RAS), regulates cardiovascular functions and exerts beneficial effects in cardiovascular disease. However, the association between HMGB1 and ACE2 has not been studied. We hypothesized th… Show more

“…In the current study, ubiquitous overexpression of ACE2 greatly elevated ACE2 mRNA and ACE2 activity in the brain; however, a recent study from our group determined ACE2 KI mice also have elevated ACE2 mRNA and ACE2 activity in peripheral tissues and plasma, respectively (Qi et al, 2015). Therefore, our subsequent experiments evaluated whether the suppressed anxiety-like behavior observed with augmented ACE2 was centrally mediated.…”

“…When a Cre-recombinase expressing mouse is bred with these animals ACE2 will be activated in the cells expressing the Cre-recombinase. Expression of Crerecombinase in the germline results in ubiquitously ACE2 overexpressing mice [58,59].…”

“…In rats, treatment with DIZE improved the myocardial infarction through inhibiting cardiac inflammation and apoptosis (Qi et al ., ). In another study, DIZE prevented hypoxia‐induced death of cardiomyocytes through inhibiting high‐mobility group box 1 protein (Qi et al ., ). There is also evidence that DIZE is beneficial in several diabetes‐induced pathologies.…”
