Angiostatic activities of medroxyprogesterone acetate and its analogues

Abstract: The effects of medroxyprogesterone acetate (MPA) (I) and related compounds (II-VI) upon angiogenesis induced by basic fibroblast growth factor (bFGF) or transforming growth factor-alpha (TGF-alpha) were investigated using a rabbit corneal system for assay of angiogenesis. Dexamethasone (Dex) was used as a positive control. The MPA analogues tested were 6,6'-dehydro-MPA (II), megestrol acetate (III), 1-dehydromegestrol acetate (IV), melengestrol acetate (V), and 1-dehydromelengestrol acetate (VI). The inhibitor… Show more

“…In contrast, high doses of oral progesterone reduce the endometrial microvascular density [9]. A direct anti-angiogenic action has been detected in assays with medroxyprogesterone acetate [10], which seems to have an angiostatic action [11].…”

“…Investigators have reported that androgens may significantly alter the development of allergic conjunctivitis in rabbits5 and that these hormones may have a beneficial influence on the corneal epithelium, acting to correct defects, facilitate wound healing,6 7 47 suppress angiogenesis,48 and stimulate mitosis 15. In fact, a pharmaceutical firm in South America (Laboratórios Frumtost SA/Allergan-Lok, Guarulhos, Brazil) has marketed topical androgens for the therapy of corneal trauma, cicatrisation, erosions, ulcers, and atrophy, as well as for promoting postoperative care after corneal transplantation.…”

“…There are several hypotheses about the effect of progestagens in the treatment for SIPS. Progestagens appear to have angiostatic activities (Yamamoto et al 1994). In addition, Arbeiter (1975) suggested that the anti-oestrogen effect of progestagens inhibits haemorrhage resulting from increased capillary permeability in the endometrium (Arbeiter 1975).…”
