Protocols were developed for the generation of haploid and doubled haploid plants from isolated microspores of carrot (Daucus carota L.). Forty-seven carrot accessions, including six inbred lines, 11 cultivars, 20 F 1 s, two BC 1 F 1 s, four F 2 s, one F 3 , and three F 4 s, were screened to evaluate the genotype influence on isolated microspore embryogenesis over 4 years. Twenty-eight accessions responded by producing embryos and/or calli. A cytological analysis showed that two modes of carrot microspore embryogenesis exist: an indirect route via calli (C mode), and a direct route via embryos (E mode). Eleven accessions were in the C mode, and 17 were in both modes. The highest production rates were in 10Y25 (a European Nantes cultivar) with 27 calli and 307 embryos, and 100Q6 (a semi-Nantes F 1 hybrid) with 176 calli and 114 embryos. The time period to produce embryos or calli differed significantly between 2 and 6 months. Cold and heat pretreatment generally had a negative impact on the induction of microspore embryogenesis, but a short pretreatment showed a positive influence on some accessions. Twentyeight lines regenerated plants from the primary individual embryos or calli of three accessions were established to analyze the ploidy level. The percentage of spontaneous diploidization showed very wide differences among the accessions and lines. Differences in leaf color intensity, leaf size, and leaf dissection were found among haploid, doubled haploid, and triploid plants.

Haploidisation is a biotechnological method used to obtain plants with improved traits that are of use to humans. Lettuce (Lactuca sativa L.), a well-known and popular leafy vegetable, is consumed worldwide. Its haploid form would provide a good basis for producing a pure line of plants (doubled haploids) allowing new varieties to be regenerated. The main aim of this work was to develop an effective haploidisation method for this economically important species. In order to stimulate the development of haploid embryos of lettuce based on our previous experience, we conducted in vivo distant pollination with fresh pollen grains of Helianthus annuus L. or H. tuberosus L. Because the haploid proembryos obtained after pollination did not develop further (despite the presence of cellular endosperm), we implemented the technique of in vitro culture of an isolated embryo sacs (surrounded by endothelium) with parthenogenetic embryos on various, modified Murashige and Skoog media. During the in vitro culture, we observed the formation of callus tissue and, after subsequent cultures of calluses, 23 haploid L. sativa plants were regenerated. The haploid status of the regenerated plantlets was confirmed by estimation of the genome size by flow cytometry, chromosome counting in root tips, stomata cell size and by disturbances in pollen formation resulting from abnormal microsporogenesis. This paper contains the complete protocol for obtaining haploid plants of L. sativa.

IntroductionIn general, haploid cells or individuals are those in which the original chromosome number has reduced by half. According to Seguí-Simarro (2010), androgenesis can be defined as a set of biological processes leading to an individual that genetically originated exclusively from a male nucleus. After spontaneous or induced genome doubling, double haploids (DHs) that are completely homozygous are obtained. The production of haploid plants can be induced by methods such as parthenogenesis, interspecific hybridization, pollination with irradiated pollen, hormonal and/or chemical treatment, and application of high-temperature stress (Forster et al., 2007;Murovec and Bohanec, 2013). In anther culture, haploids and DHs occur through direct microspore embryogenesis. This technique has no analogue for fast creation of pure homozygous lines in a single step. In pepper-plant breeding, for example, this method has helped in the development of important new varieties with improved adaptability, productivity, and disease and pest tolerance (Chunling and Baojun, 1995;Venczel and Mitykó, 1995;Arnedo Andrés et al., 2004;Nervo et al., 2007).
