And Akt‐ion! IQGAP1 in control of signaling pathways

Abstract: ERK signaling and Akt signaling are inversely correlated in some cancers. Yet, the precise molecular mechanism for crossinhibition remains unclear. In this issue of The EMBO Journal, Pan et al (2017) show that when Akt is on, its phosphorylated cytoplasmic substrate FOXO1 turns off ERK activity by reshaping the Ras-ERK scaffold IQGAP1.

“…NKG2D stimulation results in the activation of critical signaling pathways that lead to cytokine gene transcription in NK cells ( 3 ). IQGAP1 regulates multiple kinase signaling programs known to be activated in response to NKG2D ligation, including Erk1/2 and Akt ( 14 , 65 ). Therefore, we examined the requirement of IQGAP1 in NKG2D-mediated signaling in the context of cytokine production.…”

“…IQ domain-containing GTPase-activating protein 1 is a ubiquitously expressed, 190-kDa scaffold protein ( 13 ) that regulates cellular signaling in multiple contexts ( 9 , 14 , 15 ). The scaffold function of IQGAP1 is mediated through six functional domains that facilitate its interaction with various proteins and signaling molecules.…”

“…Scaffold proteins for the mitogen-activated protein kinase (MAPK) cascade were among the first to be discovered (Printen et al, 1994, Therrien et al, 1996). The expanding group of MAPK scaffolds comprises several scaffolds for the extracellular signal-regulated kinase (ERK) pathway (Choi et al, 2017), such as kinase suppressor of Ras 1 (KSR1), paxillin, MEK partner 1 (MP1), caveolin-1, and IQGAP1 (Kolch, 2005, Wortzel et al, 2011). Reported target proteins of IQGAP1 are actin, Ca 2+ /calmodulin, E-cadherin, epidermal growth factor receptor, B-Raf, MAPK/ERK kinase1/2, ERK2, and Cdc42 (Pan et al, 2012).…”

“…The domains in IQGAP1 are, starting from the N-terminus, the calponin homology domain (CHD), the poly-proline protein-protein domain (WW), the 4 IQ domains and the RasGAP-related domain (GRD) (Figure 1a). In cells, IQGAP1 functions as a scaffold protein, playing a key role in both the PI3K-Akt and Ras-ERK pathways 8 . Crucial components of these pathways are assembled by IQGAP1.…”
