Anaphylaxis due to liposomal amphotericin (AmBisome)

Abstract: Letters to the Editor study population (12.75%). Thus we can confirm that the presence of vaginal and cervical ulcers may be considered an additional risk factor for HIV transmission. Although our study was performed on sera belonging to women considered at risk for HIV infections, only 2% of them were certainly infected by HIV-1 and none was infected by HIV-2. This percentage is consistent with another study on an analogous sample of 100 women in February 1993 (data not shown). During the last 10 years, the w… Show more

“…Several clinically approved formulations, namely, liposomal doxorubicin (Doxil), liposomal amphotericin (AmBisome), and intravenous paclitaxel (Taxol), , are inflammatory through the activation of the complement system of proteins . Such immune activation may lead to a severe or even fatal condition called complement activation-related pseudoallergy (CARPA). , CARPA is used to describe hypersensitivity reactions to nanomedicines occurring in up to 45% of individuals .…”

“…73 However, the effect on clotting time was not found to be physiologically significant (prolongation ≥ 2-fold vs untreated control is considered physiologically significant). Several clinically approved formulations, namely, liposomal doxorubicin (Doxil), 57 liposomal amphotericin (AmBisome), 74 and intravenous paclitaxel (Taxol), 75,76 are inflammatory through the activation of the complement system of proteins. 77 Such immune activation may lead to a severe or even fatal condition called complement activation-related pseudoallergy (CARPA).…”

“…[24][25][26][27][28] Allergic reactions and anaphylaxis have been reported with amphotericin B deoxycholate as well as lipid products, thus justifying the recommendation of a l-mg test dose.
