Anandamide‐induced endoplasmic reticulum stress and apoptosis are mediated by oxidative stress in non‐melanoma skin cancer: Receptor‐independent endocannabinoid signaling

Abstract: Endocannabinoids are neuromodulatory lipids that regulate central and peripheral physiological functions. Endocannabinoids have emerged as effective antitumor drugs due to their ability to induce apoptosis in various cancer studies. The G-protein coupled cannabinoid receptors (CB1 and CB2) and the TRPV1 ion channel were reported to mediate the antiproliferative activity of endocannabinoids. However, receptor-independent effects also account for their activity. Our previous studies showed that the antiprolifera… Show more

“…Concerning eCBs, they generally exert tumor-suppressive effect, based on clinical observations [272]. In vitro, AEA was demonstrated to decrease invasion and metastasis by upregulating a tissue inhibitor of matrix metalloproteinases-1 (TIMP-1) [273] and to induce apoptosis via oxidative stress in COX-2- [274] and TRPV1-dependent manner [275]. 2-AG (20 mg/kg/day) showed similar properties in the rodent model, at the same time suppressing tumor immune-mediated surveillance [276].…”

“…We believe these findings could be on major importance for better understanding pathophysiology of psoriasis, because CB2 receptor activation is crucial signal for anti-oxidative and anti-inflammatory properties of endocannabinoids, due to their ability to decrease ROS as well as TNF-α, IFN-γ and other inflammatory cytokines [10]. On the other hand, enhanced CB1 receptor expression may increase oxidative stress and inflammation, which may be related to PsA as the advanced type of psoriasis [11]. It is also known that CB2 receptor expression is higher than is expression of CB1 in the immune-competent cells, which suggest that CB2 effects dominate in immune cells, including granulocytes [10].…”
