Anammox Bacterial Abundance, Activity, and Contribution in Riparian Sediments of the Pearl River Estuary

Abstract: The hypothesis of an anammox hotspot in river riparian zones was put forward based on our investigation on freshwater ecotones for over 25 years and previous anammox research. Here we used a complementary array of methods including isotope-pairing technique, quantitative PCR assays, and 16S rRNA and hydrazine synthase gene (hzsB) clone libraries to document the spatiotemporal evidence for a high abundance zone of anammox bacteria in river riparian sediment with observed abundance of 1.3-12 × 10(6) (summer) and… Show more

“…Despite these findings suggesting the functional importance of anammox, the contributions to N 2 production from denitrification were approximately 10e150 times of that from anammox in the BH and LZ columns, and denitrification was responsible for all N 2 emission in the TY column and field-fresh soil. Similar trends are also observed in other paddy soils (Zhu et al, 2011b;Yang et al, 2015), freshwater sediments (Zhu et al, 2011a(Zhu et al, , 2013Wang et al, 2012a) and mangrove sediments (Fernandes et al, 2012), suggesting a more dominant contribution of denitrification than anammox to N 2 production in terrestrial ecosystems. In contrast, the contribution of anammox to N 2 production was reported to exceed denitrification in marine ecosystems (Thamdrup and Dalsgaard, 2002;Engstr€ om et al, 2005).…”

“…Anammox has been reported to account for Ͼ80% of total N 2 production in the eastern tropical South Pacific oxygen minimum zone (OMZ) off northern Chile (5). Anammox bacteria were detected with broad biogeographic distribution in many naturally occurring anoxic environments, including aquatic ecosystems (6)(7)(8)(9) and terrestrial ecosystems (10)(11)(12)(13)(14)(15). However, their role related to activity in agricultural ecosystems remains poorly understood.…”

“…The anammox genotype is associated with hydrazine synthase, which consists of three subunits encoded by the genes hzsA, hzsB, and hzsC (21-23), re-sponsible for the synthesis of hydrazine from nitric oxide and ammonium (22). The hzsB gene is a newly developed anammox phylomarker which has been successfully applied in the investigation of the biodiversity and quantitative analyses of anammox bacteria in various environments (9,24,25); hence, hzsB was used in this study.…”

“…As the rice rhizosphere is highly oxygenated compared to bulk soil, nitrite, which is a substrate for anammox, may be produced through both denitrification and nitrification (Lam et al, 2007;Zhu et al, 2011a;Wang et al, 2012a). In oxygenlimited bulk soil with low C/N ratio, where nitrification is inhibited, denitrifiers may reduce limited nitrate diffused from rhizosphere, to nitrite (Trimmer and Nicholls, 2009), then provide anammox with nitrite.…”

“…About~3.5 g soil was transferred into a 12 ml gastight vial (Exetainer, Labco, High Wycombe, Buckinghamshire, UK), which were then filled totally with N 2 -purged overlaying water (Wang et al, 2012a) and sealed with a gastight lid. The resulting slurries were pre-incubated for 1 day to remove residual NO x À and oxygen according to our preliminary experiment when both NO 2 À and NO 3 À were under the detection limit of ion chromatograph (0.05e0.1 ppm and 0.075e0.1 ppm for NO 2 À and NO 3 À , respectively).…”
