Momordica species (Family Cucurbitaceae) are cultivated throughout the world for their edible fruits, leaves, shoots and seeds. Among the species of the genus Momordica, there are three selected species that are used as vegetable, and for medicinal purposes, Momordica charantia L (Bitter melon), Momordica foetida Schumach (Bitter cucumber) and Momordica balsamina L (African pumpkin). The fruits and leaves of these Momordica species are rich in primary and secondary metabolites such as proteins, fibers, minerals (calcium, iron, magnesium, zinc), β-carotene, foliate, ascorbic acid, among others. The extracts from Momordica species are used for the treatment of a variety of diseases and ailments in traditional medicine. Momordica species extracts are reputed to possess anti-diabetic, anti-microbial, anthelmintic bioactivity, abortifacient, anti-bacterial, anti-viral, and play chemo-preventive functions. In this review we summarize the biochemical, nutritional, and medicinal values of three Momordica species (M. charantia, M. foetida and M. balsamina) as promising and innovative sources of natural bioactive compounds for future pharmaceutical usage.

In recent years, medicinal plants have gained much attention as potential source of bioactives for the development of novel herbal drugs for primary healthcare. However, phytotherapeutic mechanisms of action of phytomedicines need to be explored comprehensively. Out of the available strategies for the said purpose, metabolomics is one, which results in the production of inclusive metabolite profiles providing a clear understanding of diagnostic changes in the levels of metabolites, leading to therapeutic monitoring of drug targets through the elucidation of metabolomic pathways. On the other hand, proteomics is also a powerful strategy to deal with systematic protein expression analysis and analyze different biomarkers compared to metabolomics during drug treatment. Currently, 1 H nuclear magnetic resonance spectroscopy, mass spectrometry (MS), Fourier transform infrared spectroscopy, gas chromatography equipped with mass spectrometry, liquid chromatography coupled with MS and ultra-performance liquid chromatography-MS coupled with multivariate statistical techniques, such as principle component analysis, partial least square and orthogonalpartial least square are among the widely applicable analytical tools for metabolite profiling, whereas two-dimensional electrophoresis and matrix-assisted laser desorption ionization time-of-flight-MS are the most accepted analytical methods for proteomic biomarker investigation. The present review summarizes the recent developments and perspectives of the biomarker investigation strategies so as to elaborate their imperative role for sustainable herbal drug developments.

Opioids are the drugs most commonly detected in overdose deaths and the second most consumed worldwide. An analytical methodology has been optimized and fully validated for the determination of codeine, morphine, 6-acetylmorphine, 6-acetylcodeine, oxycodone, oxymorphone and fentanyl in whole blood and pericardial fluid. The internal standards used were codeine-d3, morphine-d3, 6-acetylmorphine-d3 and fentanyl-d5. Before solid phase extraction, volumes of 250 μL of blood and pericardial fluid were subjected to a protein precipitation (with 750 μL of ice-cold acetonitrile) and a microwave induced oximation was performed using a solution of 1% aqueous hydroxylamine hydrochloride in PBS (1:2, v/v). Finally, the dried extracts were further derivatized with a solution of MSTFA+5% TMCS under microwave irradiation. The chromatographic analysis was carried out using gas chromatography–mass spectrometry operating in electron impact and selected ion monitoring mode. For all analytes the method was linear between 5 and 1000 ng/mL with determination coefficients (r2) higher than 0.99. Depending on the analyte and matrix the limit of detection varies between 3 and 4 ng/mL. Intra- and intermediate precision (<20%) and bias (±20%) were acceptable for all analytes in both matrices. The stability of the substances in the studied matrices was guaranteed, at least, 24 h in the autosampler, 4 h at room temperature and 30 days after three freeze/thaw cycles. This methodology was applied to real samples from the Laboratory of Chemistry and Forensic Toxicology, Centre Branch, of the National Institute of Legal Medicine and Forensic Sciences, Portugal.
