SUMMARYAmong the more than 5000 chemicals reported in cigarette smoke condensate (CSC), heterocyclic aromatic amines (HAAs) are considered to be a contributor to observed biological activity. HAAs are non-volatile and are reported at ppb levels in CSC. A new method for HAA analysis at the trace level is reported here. N, O-Bis(trimethylsilyl)trifluoroacetamide (BSTFA) containing 1% trimethylchlorosilane was employed to derivatize amino groups by heating the reagent containing a sample of CSC at 80 °C for 30 min followed by analysis employing gas chromatography-mass spectroscopy (GC-MS) in the selected-ion-monitoring (SIM) mode. This derivatization method afforded symmetrical peak shapes on a ZB-50 stationary phase and achieved instrumental limits of quantification (LOQ) at 10:1 S/N from -1 ng/mL for AαC to120 ng/mL for Glu-P-1. The chemical identity of each derivative was confirmed by comparison of retention time and mass spectra of standards. The latter were characterized by the following ions:, and m/z 73 (i.e., trimethylsilyl). CSC and its base sub-fractions were studied using the GC-MS method. Ten HAAs were screened and five were quantified in cigarette smoke condensate, while 2-5 HAAs were quantified in each of three base sub-fractions. . HETEROCYKLICZNE AMINY AROMATYCZNE JAKO ZAGROŻENIE CHEMICZNE W PRODUKTACH MIĘSNYCH PODDAWANYCH OBRÓBCE TERMICZNEJS t r e s z c z e n i e Niektóre sposoby przetwarzania żywności mogą powodować powstawanie w niej szkodliwych związków chemicznych, w tym mutagennych i kancerogennych heterocyklicznych amin aromatycznych (HCA). Substancje te tworzą się podczas obróbki termicznej produktów o dużej zawartości białka, przy czym ich ilość i rodzaj są warunkowane głównie temperaturą, czasem i rodzajem tego procesu. W niniejszej pracy przedstawiono ogólną charakterystykę oraz podział HCA, z uwzględnieniem ich budowy chemicznej i warunków powstawania. Omówiono również metody obróbki termicznej mięsa pod względem tworzenia się heterocyklicznych amin aromatycznych oraz hamowania procesów ich powstawania. Istnieje bowiem związek pomiędzy częstym spożywaniem przetworzonego mięsa a częstotliwością występowania nowotworów u ludzi. Ostatnia cześć pracy stanowi przegląd technik analitycznych oczyszczania i ekstrakcji próbek zawierających wymienione związki, a także metod ich analizy jakościowej i ilościowej.Słowa kluczowe: heterocykliczne aminy aromatyczne (HCA), obróbka termiczna żywności, przetwory mięsne, związki mutagenne, związki kancerogenne WprowadzenieDane epidemiologiczne wskazują na wyraźny związek pomiędzy niektórymi składnikami żywności a występowaniem określonych chorób, np. nowotworowych [4]. Do najbardziej szkodliwych związków wyizolowanych z artykułów spożywczych należą m.in. heterocykliczne aminy aromatyczne (HCA), wielopierścieniowe węglowo-dory aromatyczne (WWA), nitrozoaminy oraz niektóre produkty reakcji Maillarda [1,34,41]. Związki te mogą powstawać podczas przygotowywania żywności bądź jej przechowywania. Heterocykliczne aminy aromatyczne, będące przedmiotem tej pracy, Dr J. Majcherczyk, prof. dr hab. inż. K. The fundamental aromatic hydrocarbon benzene, when substituted by one or more functional groups of the amine, nitro, or halogen classes, alone or in various combinations, provides an enormous number of compounds of immense practical value, ubiquitous presence in the industrial world, and often heightened toxicity relative to benzene itself, a notorious cause of blood cancer. This chapter begins by providing a general overview of aromatic compounds possessing amino and/or nitro substituents, with or without halogen substituents. The production and uses, exposure and biomonitoring, toxicity, and toxic mechanism of action are introduced. The remainder of the chapter focuses on specific compounds in a roughly systematic order based upon chemical structure, expanding on the same subheadings as covered in the general overview. The nomenclature of these compounds can be very confusing since many have traditional names that sometimes markedly differ from their systematized formal names. In this chapter, the same traditional names are used as were used in the previous edition of this publication, but if there is ambiguity of nomenclature, the Chemical Abstracts Service (CAS) numbers provided herein correspond to unique structures, each of which may have several names depending upon the naming convention. In general, when an aromatic compound has an amino or a nitro substituent, it is referred to as an “aniline” or a “nitrobenzene,” respectively, based upon the names of the parent compounds, even though other substituents may be present. When amino and nitro groups are present on the same benzene ring, “aniline” takes precedence, and both “aniline” and “nitrobenzene” take precedence over halogens. There are many exceptions to these general rules, however.
