Analysis of urine for cysteine, cysteinylglycine, and homocysteine by high-performance liquid chromatography

Abstract: A new analytical method is proposed for simultaneous determination, by liquid chromatography, of the three main urinary thiols-cysteine, cysteinylglycine, and homocysteine. To measure the total amount of these thiols urine is reduced with sodium borohydride, to convert disulfides to thiols which are then derivatized with 2-chloro-1-methylquinolinium tetrafluoroborate. Separation and quantitation of the 2-S-quinolinium thiol derivatives formed were achieved by high-performance liquid chromatography with detecti… Show more

“…Average total CysGly content in plasma and urine was 29.99 ± 7.98 and 2.09 ± 1.26 μmol L −1 , respectively. These results are consistent with those reported earlier [9,20,25,26], and this indicates that elaborated assay can be successfully used in large populations. It is safe for the operator and environmentally friendly, meeting the needs of the biological sample analysis.…”

“…The integration windows for each of the scan methods are shown in Table 1, and the LODs of The absolute LODs for the thiols were in the sub-picomole range, and they were better than the LC-UV methods (11,12,33), most of the MS based detection methods (5,(34)(35)(36)(37)(38)(39), and the results via fluorescence measurement (40,41).…”

“…1,5 Thus, pursuing an a sensitive, reliable, and convenient approach for trace analyzing L-cysteine is of great significance in biological, medical, and clinical studies and attract considerable interest in recent years. Various methods have been reported for the determination of L-cysteine, such as flow injection, 6 electrochemistry, 7-13 high-performance liquid chromatography, 14,15 and capillary zone electrophoresis.…”
