Analysis of the Resistome of a Multidrug-Resistant NDM-1-Producing Escherichia coli Strain by High-Throughput Genome Sequencing

Abstract: The resistome of the multidrug-resistant Escherichia coli strain 271 carrying the plasmid-mediated bla NDM-1 carbapenemase gene was analyzed by high-throughput genome sequencing. The p271A plasmid carrying the bla NDM-1 gene was 35.9 kb in size and possessed an IncN-type backbone that harbored a novel replicase gene. Acquisition of the bla NDM-1 gene on plasmid p271A had been likely the result of a cointegration event involving the transposase of Tn5403. The expression of bla NDM-1 was associated with the inse… Show more

“…This may be related to both the presence of specific alleles which might have different inactivation levels, by the concomitant presence of unidentified macrolide-resistance mechanisms or by mph(A) gene expression levels. In the latter respect, the presence of a series of mph(A) gene copies has been described in the same genetic structure (Poirel et al, 2011).…”

“…This may be related to the presence of specific alleles which might have different inactivation levels, with the presence of unidentified macrolideresistance mechanisms or with the gene expression levels. In this latter line, different TMMR or multiple copies of the same TMMR may be present in the same or in different genetic structures within a microorganism (Baker et al, 2015;Katayama et al, 1998;Poirel et al, 2011;Soge et al, 2006). Moreover the final copy numbers of a specific gene also depend on its precise location (e.g.…”

“…The bla NDM-1 gene was originally identified among Klebsiella pneumoniae isolates from infections originating in India. However, it was quickly realized that this gene was plasmid borne, was present on multiple plasmid types ( [164][165][166][167], and was rapidly spreading between bacteria (168). The first report of bla NDM in Salmonella involved a patient with a Klebsiella pneumoniae infection that was resistant to carbapenems (169).…”

“…This type of plasmid was previously known to be involved in the transmission of VIM-1, KPC-2, and CTX-M-1 as well as NDM-1 among K. pneumoniae isolates and has been also prevalent in E. coli and Salmonella species (1). Various replicon types are involved in bla NDM-1 -carrying plasmids (19), but IncN has recently been found in only one NDM-1-positive E. coli strain (18). Highly efficient transmission of bla NDM-1 -carrying plasmids (21) may explain the diversity and worldwide spread of blaNDM-1-carrying Enterobacteriaceae.…”
