Analysis of the Gonadal Sex of Five Intersex Pigs Using Y Chromosomal Markers

Abstract: DNA isolated from gonadal tissue of five intersex pigs was analyzed for the presence of the Y chromosomal markers ZFY and DYZI. Four cases showed no detectable male-specific DNA-sequences. The fifth case exhibited a weak male-specific band at Southern blot analysis with the human ZFY probe and showed a male-specific PCR-product with DYZl primers, indicating an XXjXY gonadal chimera.

“…true hermaphroditism, male pseudohermaphroditism, female pseudohermaphroditism and males with or without sexual ambiguities). In some of these cases, complementary molecular analyses revealed the lack of specific Y chromosome sequences (including the SRY gene) [Thomsen and Poulsen, 1993;Pailhoux et al, 1994Switonski et al, 2002]. Female karyotypes and lack of the SRY gene were also observed in other species as illustrated in this article.…”

“…The same incidence has been reported in XX sex reversal mice, but this translocation has not yet been identified in dogs or pigs. In these latter species, all XX sex reversal cases have been SRY negative (Thomsen and Poulsen ; Meyers‐Wallen et al. ; Meyers‐Wallen ).…”
