Analysis of the expression of platelet antigens CD9 and CD41/61 in transgenic rabbits with the integrated human blood clotting factor VIII gene construct

Abstract: Abstract. The objective of this research was to study the expression of cell membrane molecules CD9 and CD41/61 of transgenic rabbit with integrated human factor VIII (rhFVIII) gene construct. The expressions of these molecules have been monitored during two lactations of transgenic rabbits and simultaneously compared with the expression of the same molecules of non-transgenic rabbits. The immunochemical analysis by indirect immunofluorescence, ELISA and indirect immunoperoxidase staining of blood cells and ud… Show more

“…This has resulted in a multitude of new strains to address interesting research questions. Strains have also been developed that express human recombinant proteins in rabbit milk (Chrenek et al, 2007;Dragin et al, 2005;Hiripi et al, 2010;Houser et al, 2010;Lipinski et al, 2012;Simon et al, 2011;Soler et al, 2005). “…These human proteins have resulted in antigen production for rotavirus vaccine creation, human factor VIII that could be used to treat hemophilia (Chrenek et al, 2007;Krylov et al, 2008;Simon et al, 2011) and human growth hormone that could supplement a deficiency in that hormone (Lipinski et al, 2012). Rabbits that express enhanced green fluorescent protein (EGFP) in various tissues have been created for the purpose tracking cells, which is important for tissue engineering and regenerative medicine studies Takahashi et al, 2007;Yin et al, 2013).…”

“…Auto‐Abs against this GP is produced in ITP, which leads to reduced half‐life of platelets and thrombocytopenia in these patients . The expression of CD41/CD61 on the surface of activated platelets results in the increased expression of CD9, a surface GP of human platelets involved in the activation and adhesion functions of platelets . Therefore, we can say that the evaluation of changing expression of CD41/CD61 can determine the function of platelets in ITP patients.…”

“…One such obstacle is how to increase the quantity of target gene expression. Therefore, optimized expression vectors are needed to increase transgene expression in the mammary gland (Agnew and Pfleger, 2011; Pipe et al, 2011; Simon et al, 2011).…”
