Analysis of the Anti-Inflammatory and Analgesic Mechanism of Shiyifang Vinum Based on Network Pharmacology

Abstract: Objective. The possible core active compounds and potential mechanism of action of Shiyifang Vinum were explored through network pharmacology and in vitro enzyme activity verification experiments. Methods. We screened the core active components and the action targets of Shiyifang Vinum through the TCMSP database and literature mining and drew a Venn map of the intersection with anti-inflammatory and analgesic-related gene targets. Go and KEGG analyses were enriched with the David database. The compound target … Show more

“…To analyze the biological pathways of genes in the PPI network ( 43 , 46 ), the computational R-language package of “clusterProfiler ( 47 )” (version 4.1.0) was applied to analyze Gene Ontology (GO) enrichment in biological function/process (BP), cellular component (CC), and molecular function (MF) (adjusted to p < 0.05). The DAVID database (available online: https://david.ncifcrf.gov/home.jsp , last updated in May 2016) ( 48 ) was used to analyze Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment with the “Homo sapiens” setting (adjusted to p < 0.05).…”

“…The active ingredient andrographolide extracted from this herb has been tested as a multitarget inflammatory drug, showing good results against multiple sclerosis, some respiratory diseases, and osteoarthritis [ 15 ]. Complex ancient preparations used for centuries, such as the Shiyifang Vinum containing 13 herbs, have been employed to treat pain and inflammation [ 16 ]. One of the first anti-inflammatory treatments described by ancient civilizations, such as Egyptians, Greeks, Sumerians and Chinese, was the use of the willow bark [ 17 , 18 , 19 ].…”
