BackgroundThe pathology of osteoarthritis (OA) is partly attributed to genetic factors; however, the role of ADAM12 polymorphism is still controversial. It is necessary to perform a meta-analysis to investigate this possible correlation.MethodsCase–control studies on the association between OA susceptibility and ADAM12 polymorphism were comprehensively collected by searching PubMed, Embase, and Web of Science. Odds ratios (ORs) and 95% confidence intervals (CIs) were pooled to evaluate OA risk that was possibly conferred by ADAM12 variant. The analyses were performed not only among general population but also in male and female groups.ResultsA total of 8 studies with 10 populations were finally included in this meta-analysis. In the general population, 4 comparisons were carried out (C allele vs G allele, CC vs GG, GC + CC vs GG, and CC vs GC + GG) and found that ADAM12 rs3740199 polymorphism was not associated with increased OA vulnerability. On the other hand, the analyses stratified by gender made 5 comparisons (C allele vs G allele, CC vs GG, GC vs GG, GC + CC vs GG, and CC vs GC + GG). It was shown that rs3740199 polymorphism (GC + CC vs GG) was a risk factor for OA among male patients (OR =1.45, 95% CI =1.04–2.01). Sensitivity analysis indicated that it was an unstable outcome. No correlation was identified in women. Neither heterogeneity nor publication bias was detected in the analyses mentioned above.ConclusionADAM12 rs3740199 polymorphism is likely to be associated with OA susceptibility among male patients, other than the general population. More studies are needed to confirm this observation. The mechanism by which ADAM12 variant plays a role in OA pathogenesis is also warranted and important for interpreting this possible correlation.

Objectives: This study aims to investigate the association between a disintegrin and metalloproteinase 12 polymorphisms and susceptibility to knee osteoarthritis (OA) through a meta-analysis involving only studies that present original data. Materials and methods: A meta-analysis was performed on the association between a disintegrin and metalloproteinase 12 rs3740199, rs1871054, rs1278279 and rs1044122 polymorphisms and knee OA. An electronic search of all relevant studies published up to January 2017 was conducted using MEDLINE and EMBASE. The study included six case-control studies including 2,966 patients (850 males, 2,116 females) with knee OA and 4,901 controls (2,286 males, 2,615 females). Results: No significant association was found between the rs3740199, rs1278279, and rs1044122 polymorphisms and knee OA when all studies were pooled into the meta-analysis. However, a significant association was found between the rs1871054 polymorphism and the knee OA risk (allele contrast: odds ratio (OR)=1.42, 95% confidence interval (CI)=1.02-1.98, p=0.04; dominant: OR=1.36, 95% CI=1.02-1.82, p=0.04; recessive: OR=1.83, 95% CI=1.14-2.94, p=0.01; and addictive model: OR=1.85, 95% CI=1.06-3.23, p=0.03). In the subgroup analysis, a significant association was found between the recessive (OR=0.44, 95% CI=0.21-0.93, p=0.03) and addictive model (OR=0.29, 95% CI=0.12-0.69, p<0.01) in the male group. Conclusion: Our meta-analysis suggests that the rs1871054 polymorphism is associated with susceptibility to knee OA and that the GG genotype of rs3740199 was associated with knee OA in the male group. Chondrocyte exposure to inflammatory stimuli in several arthritic conditions, including osteoarthritis, results in the well-characterized induction of extracellular matrix degrading proteinases, notably members of a disintegrin and metalloproteinase (ADAM) with thrombospondin domains and matrix metalloproteinase families. Here we briefly review the less-studied ADAM family of proteinases in chondrocyte and cartilage biology. Following damage, cartilage is exposed to neurovascular peptides, and in this study we hypothesized that substance P and bradykinin, alongside inflammatory cytokines, may modulate chondrocyte steady-state messenger RNA levels for the proteolytic ADAM family members as well as for key cytokines and neuropeptides. We compared chondrocytes cultured in both two-dimensional and three-dimensional (3D) environments and found that 3D culture generally resulted in repression of expression of the genes under investigation, with the exception of anti-inflammatory interleukin 10 which was markedly upregulated in a 3D environment. Substance P and bradykinin had little effect on ADAM family expression, but further investigation revealed that a combination of bradykinin and cytokines led to enhanced expression of ADAM28 and a synergistic upregulation of interleukin 6, also observed under hypoxic conditions. Overall these data reveal wider chondrocyte responses to neurovascular peptides which may have an impact in an osteoarthritis context.
