Tuberculosis (TB) is an ancient human disease caused by Mycobacterium tuberculosis which affects the lungs, making pulmonary disease the most common presentation. Herbal medicine began to be developed as hepatoprotection. The hepatoprotection effects of Plantago major extract on serum glutamic pyruvic transaminase (SGPT), liver tissue Malondialdehyde (MDA), and histopathological changes were evaluated in rifampicin and isoniazid-induced hepatitis rats. Thirty male Wistar rats were divided into 6 groups: Group I received 1 % PGA, with hepatitis induced in the remaining groups by rifampicin and isoniazid 50 mg/kg BW/day, group II was only given rifampicin and isoniazid, group III was given curcuma 10.8 mg/kg BW/day and groups IV (PM1), V (PM2), and VI (PM3) were given Plantago major extract at a dose of 20.3, 40.5, and 81 mg/kg BW/day, respectively. The rats were treated for 28 days. Administration of Plantago major extract (20.3 and 40.5 mg/kg BW/day) inhibited the elevation of serum SGPT and MDA levels, with less portal inflammation than the negative control group. The rats treated with the higher dose of 81 mg/kg BW/day had serum SGPT, MDA, and percentage portal inflammation equivalent to the negative control group. The Plantago major extract at a dose of 20.3 and 40.5 mg/kg BW can inhibit the elevated serum SGPT. Plantago major extract exerts dose-dependent hepatoprotection effects in a rifampicin-isoniazid induced hepatitis rat model by reducing elevated levels of SGPT, liver tissue MDA, as well as the percentage of portal inflammation. HIGHLIGHTS To the knowledge of the authors, there has been no comprehensive work dedicated to antituberculosis drugs The administration of Plantago major extract showed hepatoprotector effects, as evidenced by the inhibition of elevated levels of SGPT, MDA, and a lower percentage of portal inflammation than the negative controls Plantago major extract can be used as herbal medicine that can be juxtaposed with the main drug in TB patients GRAPHICAL ABSTRACT

Background: The evaluation of toxicity is of paramount importance in the screening of a new compound. Basella alba mucilage possesses a versatile excipient property that can be innovated with its chemical modification to get the functionalized mucilage. A few pharmacological activities of Basella alba mucilage have also been reported earlier, but its toxicity study in rats has yet to be discussed. Aim: The study aims to assess the in vivo toxicity of carboxymethylated Basella alba mucilage in Wistar albino rats for 28 days. Material and Methods: In the current investigation, Carboxymethylated Basella alba mucilage is taken, and its subchronic toxicity study is carried out in forty-eight healthy rats (twenty-four male rats and 24 female rats), divided into four groups containing six rats of each sex. All the biochemical and hematological parameters and histopathological investigation were estimated for all the animal groups. Result: The subchronic toxicity study reveals that the modified mucilage is safe for all doses (20mg/kg body weight, 40mg/kg body weight, 80 mg/kg body weight). The study showed no significant difference in the dose group's behavioral toxicity, nephrotoxicity, and hepatotoxicity compared to the control. All the hematological and biochemical parameters lay in the normal range. The histopathological examination of treatment groups showed no abnormality or lesion in the tissue samples of internal organs. Conclusion: The study confirms the safety of Carboxymethylated Basella alba mucilage for use in pharmaceutical formulations.

Background: Cleome brachycarpa, magical species full of phytochemicals, has magical medicinal properties and should be evaluated extensively. Objectives: We evaluated the anti-inflammatory and analgesic activity of Cleome brachycarpa and the effect of its extract on various hematological parameters, cholesterol levels, and liver enzymes to ensure the safe use of this natural product. Methods: Cleome brachycarpa was evaluated for its anti-inflammatory and analgesic effects at 200 mg/kg compared to diclofenic sodium and morphine, respectively. For anti-inflammatory activity, Wistar strain albino rats were pooled and divided into four groups: Negative control (normal saline), positive control (2% acetic acid), standard (Diclofenic sodium 10 mg/kg), and test (Cleome brachycarpa extract orally) groups for 10 days. For analgesic evaluation, mice were divided into control (normal saline), standard (morphine 10 mg/kg), and test (Cleome brachycarpa extract 200 mg/kg) groups and analyzed by the tail-flick method from zero to six hours. An assortment of blood parameters was evaluated, including white cells, red cells, hemoglobin level, hematocrit value, mean corpuscular volume, and mean corpuscular hemoglobin concentration. Besides, we computed the number of platelets, cholesterol, and enzyme (liver) level to ensure the safe use of this natural product. For this purpose, 60 rabbits were collected and divided into groups: Group A (control group) of 30 rabbits pooled without any treatment and Group B (treated group) of 30 rabbits receiving 200 mg/kg of Cleome brachycarpa. After 30 days, 4 mL blood sample was obtained by cardiac puncture. Results: Plethysmometer evaluation of anti-inflammatory effects showed maximum inflammatory inhibition of 29.42% at the sixth hour. Moreover, tail flick analysis showed maximum pain inhibition of 55.10% at the sixth hour, comparable to the standard drug. Furthermore, hematological data were analyzed statistically and showed insignificant results (P ≥ 0.05), indicating no prominent changes in hematological parameters, except for SGPT, a liver enzyme that increased after 30 days of treatment (P ≤ 0.05). Elevated levels of SGPT are usually reported with several drug administrations like NSAIDs and anti-TB drugs, but still, it should be further investigated. Conclusions: Cleome brachycarpa showed promising anti-inflammatory and analgesic effects without producing any potent change in enzymes except SGPT, which would be evaluated further.
