Analysis of Plasma microRNA Associated with Hemolysis

Abstract: We analyzed the effect of hemolysis on microRNA profi le of blood plasma. It was found that hemolysis of ~0.05% erythrocytes in a sample signifi cantly affected the concentration of 9 microRNA: hsa-miR-486-5p, hsa-miR-16-5p, hsa-miR-451a, hsa-miR-106a-5p, hsa-miR-17-5p, hsa-miR-93-5p, hsa-miR-20a-5p, hsa-miR-107, and hsa-miR-20b-5p. The effect of hemolysis on plasma content of miR-17 family microRNA was demonstrated.

“…Age and sex did not have effect on the miRNA expression profile, with no significantly differentially expressed miRNAs (DEmiRs), apart from one miRNA (eca-miR-8908j) that passed the lower significance threshold usually suggested for DEmiRs (upregulated in males with log 2 fold change (log2FC) = 4.23; FDR = 0.013). The absorbance at 414 nm had a significant effect on the expression of eca-miR-25 (FDR = 4.15e-4) and eca-miR-486-5p (FDR = 0.066) that was previously reported as haemolysis dependant [27]. We identified 50 differentially expressed (DE) miRNAs (42 known and 9 novel) between pony and Warmblood serum at a stringent significance threshold FDR < 0.001.…”

“…MiR-451 is a miRNA contained in red blood cells and its level is correlated with haemolysis; 14,22 it was not significantly altered by ECMO in our samples. Monitoring haemolysis, which is frequently observed during ECMO, 5 is critical when dealing with miRNAs because it is reported to be a significant source of variation in miRNA profiling.…”

“…Both prior studies employed RNU6 as a reference. While hsa-miR-451a has been found to act as a tumor suppressor [64,65], it is also a marker of hemolysis [66] and thus we suggest that care should be employed to exclude samples with hemolysis before analysis. The miRNA ratio of hsa-miR-451a and hsa-miR-23a-3p has been employed by others [56,67] to monitor for sample hemolysis.…”
