Analysis of long noncoding RNA expression profiles in the whole blood of neonates with hypoxic‐ischemic encephalopathy

Abstract: Long noncoding RNAs (lncRNAs) have been shown to participate in many biological processes. To investigate the expression profiles of lncRNAs and their potential functions in neonatal hypoxic‐ischemic encephalopathy (HIE), we detected the lncRNA and messenger RNA (mRNA) expression in the peripheral blood samples from HIE patients and controls using a microarray. A total of 376 lncRNAs and 126 mRNAs were differentially expressed between the HIE and the non‐HIE samples (fold change > 2). Quantitative real‐time po… Show more

“…neonatal Hie is one of the common causes of death or permanent disability, particularly in developing countries (24,25). The crucial role of mirnas (mirs) and long non-coding rnas in Hie pathologic processes have been previously demonstrated (19,26). The expression level of mir-374a is significantly downregulated in the umbilical cord blood of neonatal Hie (26).…”

“…The qPCR conditions were as follows: Initial denaturation at 50˚C for 2 min, 95˚C for 10 min, followed by 40 cycle at 95˚C for 15 sec and 60˚C for 30 sec. The expression of circrna was normalized to the 18S ribosomal rna, using the 2 -ΔΔcq method (19).…”

“…Studies in human neonates diagnosed with HIE and animal models have shown that lncRNAs are aberrantly expressed under hypoxic conditions and might be implicated in regulating the expression of protein-coding genes involved in pathological processes associated to HI brain damage. A microarray study in human neonates showed that neonatal HI dramatically changed the expression patterns of numerous ncRNAs in peripheral whole blood (146), including 376 lncRNAs and 126 mRNAs involved in the immune system and nervous system. A study on the P7 HI rat cortex identified 7,157 differentially expressed mRNA transcripts, and 328 differentially expressed lncRNAs targeting mRNAs involved in inflammatory and immune responses, wounding, and neurological system processes (147).…”

“…7,20 Therefore, most researchers always indirectly predict the functions of lncRNAs by performing GO and pathway analysis of their neighboring and distantly target genes. 21,22 However, to investigate the integrated functions of lncRNAs, we per- inhibition of IL-10 has been proven to recover specific CD8 + T-cell response and accelerate virus eradication in in vivo and in vitro studies. 24,27 As shown in Figure 5B, IL-10 was positively and negatively correlated with five lncRNAs and two lncRNAs, respectively, indicating that these lncRNAs might participate in the molecular mechanism of CHB through mediating the functions of IL-10.…”
