We aimed to prospectively investigate the paternal antigen-induced cytokine secretion by peripheral blood mononuclear cells (PBMC) in response to hormone treatment in women undergoing in vitro fertilization (IVF) and to examine the predictive value of the cytokine secretion in the outcome of IVF treatment in a pilot study. Twenty-five women were included and IVF treatment was successful for six and unsuccessful for 19 women. Blood samples were collected before IVF treatment, on four occasions during IVF and 4 weeks after embryo transfer. The numbers of Th1-, Th2-and Th17-associated cytokine secreting cells and cytokine levels in cell supernatants were analyzed by enzyme linked immunospot-forming (ELISpot), enzyme-linked immune-sorbent (ELISA) or Luminex assay. None of the cytokines (IFN-γ, IL-4, IL-5, IL-10, IL-12, IL-13, IL-17, TNF and GM-CSF) had any predictive value regarding IVF outcome. The majority of the cytokines reached their peak levels at ovum pickup, suggesting an enhancing influence of the hormonal stimulation. Pregnancy was associated with a high number of IL-4-, IL-5-and IL-13-secreting cells four weeks after ET. In conclusion, the results do not support our hypothesis of a more pronounced peripheral Th1 and Th17 deviation towards paternal antigens in infertile women with an unsuccessful IVF outcome, although this is based on a small number of observations. Thus, a larger study should be conducted to confirm this conclusion. Higher numbers of Th2-associated cytokinesecreting cells in pregnant women four weeks after ET do corroborate the hypothesis of a Th2 deviation during pregnancy.

A ccording to modern data, chronic endometritis is the result of imbalance between hormonal and immune systems of the body, on the one hand, and microbial pathogens on the other. The disease can develop as an outcome of inadequate treatment of acute endometritis, and as primary chronization, in women with abnormal balance between the activity of the processes of destruction, remodeling and proliferation of tissues (primary genetic defects, immunodeficiency, etc.) [8].Remodeling of the extracellular matrix one of the main mechanisms for maintaining the homeostasis in any tissue, as in norm and in pathology. The activity of matrix metalloproteinases is an important factor determining the intensity of remodeling of the extracellular matrix [4,5]. There is evidence of gelatinase B role in metastasis in oncology, in the formation and development of arthritis, paradontitis, ulcer defects of gastro-intestinal tract. Note, that performance of the activity of metalloproteinases (gelatinase, collagenases, stromelysin, etc.) changes in different ways, reflecting, in some cases, destructive processes, and initiation of cell proliferation in others [6,7]. Nowdays we have no unambiguous information about the role and importance of metalloproteinases in the pathogenesis of acute and chronic inflammation, including the pathogenesis of chronic endometritis. Insufficiently stu- Волгоградский государственный медицинский университет, Российская ФедерацияIn the study was evaluated the role of intraerythrocytic magnesium, the specific number and intensity of expression of gelatinase B positive cells on the mechanisms of chronic experimental inflammation of the endometrium in rats. Experiment was performed on 40 adult female white Wistar rats, which were divided into 3 groups. The first group (n=10) -initial state, the second group (n=10) -control series, the third group (n=10) -simulation of endometritis. On the 51st day after modeling chronic inflammation of the endometrium of the rats were receiving the blood from the inferior Vena cava and subclavian veins, the tissue of the uterine wall. Our study showed a reduction in the specific number of gelatinase В positive cells in the stroma (cellular component), the intensity of expression decreased of gelatinase-B in both the stroma and glandular epithelium. The decrease in intraerythrocytic levels of magnesium in the blood from different regions is a strong direct correlation with the number and intensity of expression gelatinase B. Thus, the decrease of intraerythrocytic magnesium, immunohistochemically activity indicators of gelatinase B on the background of the morphological picture of chronic endometritis, act as important links of the chronic endometrium inflammation. Abstract. The aim of the present study was to investigate the correlation of matrix metalloproteinase (MMP)-9 and tissue inhibitor of matrix metalloproteinase inhibitor (TIMP)-3 expression with spontaneous abortion (SA) during early pregnancy. The villus tissues of 30 SA cases and 20 requested abortion cases were collected during surgery and constituted the SA and normal abortion (NA) groups, respectively. The total villous RNA was extracted and the expression levels of MMP -9 and TIMP-3 mRNA were detected by reverse transcription-polymerase chain reaction (RT-PCR) assay to calculate the MMP-9/TIMP-3 mRNA ratio. The MMP-9 mRNA expression level and MMP-9/TIMP-3 mRNA ratio of the SA group were significantly higher than those of the NA group (P<0.01), while the TIMP-3 mRNA levels of the two groups were similar (P>0.05). The MMP-9 mRNA expression level of the SA group was higher than that of the NA group; thus, the MMP-9/TIMP-3 mRNA ratio was higher. These results suggest that the expression level of MMP-9 mRNA and the MMP-9/TIMP-3 mRNA ratio are associated with SA.
