Analysis of genetic relationships and identification of lily cultivars based on inter-simple sequence repeat markers

Abstract: ABSTRACT. Inter-simple sequence repeat (ISSR) markers were used to discriminate 62 lily cultivars of 5 hybrid series. Eight ISSR primers generated 104 bands in total, which all showed 100% polymorphism, and an average of 13 bands were amplified by each primer. Two software packages, POPGENE 1.32 and NTSYSpc 2.1, were used to analyze the data matrix. Our results showed that the observed number of alleles (N A ), effective number of alleles (N E ), Nei's genetic diversity (H), and Shannon's information index (I)… Show more

“…Results showed that primers namely ISSR 851,ISSR 22,219,ISSR 857,ISSR 861 and ISSR 22,218 that have above average PIC, MI and Rp, were to be more efficient. Previously researchers have compared the genetic diversity in other floricultural bulbous crops, with pedigree information (Patra et al 2008;Cui et al 2014;Anderson et al 2010). In the present study, genotype Arka Shobha, belonging to of some genotypes could be proved by their parental relationship.…”

“…[32] Genetic diversity parameters, such as Nei's genetic identity (I) and genetic distance (D), were estimated using POPGENE program v.1.31. [33] I reflects the genetic relationship of the two samples; that is, I = 1 or 0 indicates a close genetic relationship. D reflects the genetic variability of the samples, that is, D = 1 or 0 indicates high genetic variation.…”

“…Molecular analysis indicates that lilies arose in Eurasia, nearly 68 million years ago (Leitch et al, 2007). Lilies have been bred for more than 200 years (Peng, 2002;Cui et al, 2014), and more than 10,000 cultivars have been developed to date (Younis et al, 2014). Despite genetic similarities, species of the genus Lilium have evolved with notable phenotypic diversity, which is of considerable evolutionary significance (Patterson and Givnish, 2002).…”

“…Inter-simple sequence repeats (ISSR) are among the most useful DNA markers that have overcome most of these limitations, and have been efficiently and widely used in current studies addressing the genetic variability and diversity of plants and crops (Taheri et al, 2012;Karimi et al, 2014). At present, only a few studies have examined the diversity of Lilium species using molecular markers, including RAPD (Huang et al, 2009), ITS (Sultana et al, 2011;Du et al, 2014a) and ISSR (Guo et al, 2011;Xi et al, 2012;Zhao et al, 2014;Cui et al, 2014).…”
