Obesity is a serious global problem that is considered a great pandemic. It was related to many non-communicable diseases, including type 2 diabetes mellitus (T2DM). Crab (Crustacea: Decapoda) is one of the healthiest sea foods all over the world, being very rich in unsaturated fatty acids, especially in the female gonads and hepatopancreas. Therefore, this study was designed to explore the anti-obesity effect for the soft tissue extract of the female Nile crab Potamonautes niloticus. The chemical constituents of methanol extract from crab´s meat and soft Dichloromethane were evaluated using Liquid Chromatography with tandem mass spectrometryand Nuclear Magnetic Resonance. Also, the effect of this extract on obesity induced in zebra fish adults was studied in vivo, using real time PCR (qPCR) quantification of different lipid-metabolizing enzymes´ genes. More specifically, these analyzed transcripts were for enzymes related to de novo lipogenesis [i.e., fatty acids synthase (FASN) and acetyl- CoA carboxylase alpha (accα)]; adipogenesis [i.e. diacylglycerol O-acyltransferase 2 (dgat2)]; and lipid catabolism through β-oxidation [i.e. acetyl-CoA oxidase 1 (acox1)]. Moreover, the body contents of free cholesterol (TC) and triglycerides (TG) were assessed. The results showed that the extract contains amino acids such as cyclo-(Ala-4-hydroxy-Pro, Ile-pro, isoleucyl-proline, cyclo-(Leu-Leu), cycloleucylleucine, valyl-Leucine, L-valyl-L-phenylalanine, cyclo(L-leucyl-L-phenylalanyl), cyclo-[Tyr-(4-hydroxy-Pro)], and leucyl-phenylalanine); long chain unsaturated fatty acids such as (9-octadecenoic acid, 13-keto-9Z,11E-octadecadienoic acid, (+/-)-11-Hydroxy-5Z,8Z,12E,14Z,17Z-Eicosapentaenoic acid, and (.+/-.)-17-hydroxy-4Z,7Z,10Z,13Z,15E,19Z-docosahexaenoic acid); long chain saturated fatty acids such as (3-Hydroxyoctadecanoic Acid and 1-stearoyl-2-hydroxy-sn-glycero-3-phosphocholine); sphingolipids; fatty amide; and Monoacylglycerol. Furthermore, injection of crab extract to adult zebrafish induced a significant decrease of TG levels and the expression of the gene of the primary enzymes’ genes related to fatty acids β-oxidation, i.e., acox1. Meanwhile, crab extract injection induced significant down-regulation in the key lipogenic enzymes´ genes, i.e. accα, fasn, and dgat2. Therefore, P. niloticus soft tissue extract showed promising ingredients and capabilities that could reduce obesity through interference with lipids´ synthesis and stimulation of lipids´ degradation. Further investigations are recommended to elucidate the mechanism of action of Nile crab bioactive ingredients on obesity and other related health problems, such as T2DM.

Crabs of the genus Charybdis are some of the world’s most aggressive and voracious marine invasive crustaceans. They are found in a wide variety of ecosystems in the Red Sea. Their near ubiquity in diverse marine habitats, however, makes them a possible resource for novel marine-derived bioactive products. To provide an important insight into the potential for C. natator as a beneficial product, its shell methanolic extract was chromatographically analyzed for identification of potential bioactive ingredients. Additionally, two different doses, i.e. a low and a high dose, of this extract were tested for their ability to protect against copper-induced oxidative stress and proinflammatory response in adult zebrafish Danio rerio. Gene expression levels of transcripts of proinflammatory mediators, i.e. nf-κβ and tnf-α; antioxidant enzymes, i.e. sod, cat, and gpx; and lipid-metabolizing enzymes, i.e., acox1 and fasn, were determined. The results showed that C. natator shell is rich in potential bioactive metabolites, including diverse unsaturated fatty acids, alkanes, flavonoids, and phenolic acids. Most antioxidant and proinflammatory transcripts in the protected groups were restored to levels that were lower than those in the CuSO4-stressed group. The low dose showed special success in inducing these effects. Also, the low-dose-protected group showed significantly elevated acox1 and decreased fasn, suggesting the capability of crab shell extract at a low dose to assist lipolysis and inhibit lipogenesis. The abundant presence of saturated fatty acids in the shell extract can be the reason for the inadequacy of the high dose to promote the antioxidant and anti-inflammatory activities. These results suggest that there is a potential to develop therapeutic industrial uses for C. natator shell, instead of considering it as a discard. Its wealth of bioactive metabolites may contribute to the market for natural products to combat inflammatory and oxidative stress, the origin of many diseases in the modern world.

The rice field crabs are freshwater crabs that can be considered as pest in paddy field as well as a stable food source by local people. Either of these roles requires a good knowledge of its reproductive biology to manage population in nature. In this study, reproductive and endocrine cycles of male and female, and development of embryo were examined. Adult Esanthelphusa nani were field caught from Nan province during November 2015 to November 2016. Crabs were euthanized and dissected for reproductive tracts. Accessory organ nearby the mandibular muscle was collected for endocrine activities in male, while hemolymph was collected for vitellogenin measurement in female. Gonadosomatic index (GSI) was calculated based on the relative weight of the reproductive tract and body weight. Male GSI showed a significant temporal difference with the peak in wet season (September). Testis can be divided into 3 stages with increasing in size from 1) resting, 2) developing, and 3) active stages. Microscopic examination revealed the different activity among three stages. Meanwhile, mature sperms can be found in male reproductive tract throughout the year, indicating prolonged spermatogenic activities. Moreover, mature sperms can be deposited and found in female spermatheca. Female GSI showed two significant peaks in dry season (February) and wet season (June and August). Female GSI profile is in agreement with levels of phospholipoprotein in hemolymph. Ovaries changed in size and color due to presence of yolk and mature oocytes. Microscopic examination of ovarian tissue showed that proportions of oocyte class in ovarian tissue could be used as an important tool for identifying ovarian cycle into five stages including 1) resting, 2) vitellogenic, 3) pre-mature, 4) mature, and 5) spawning and regenerating stages. Ovigerous crabs found during March and April, 2016 were collected for embryonic study. Eggs were randomly sampled and fixed in ethanol. Eggs were initially examined and recorded for embryonic stage under a stereomicroscope. Afterward, fluorescent technique was applied to observe morphology and identify stage of development under a confocal microscope. It was found that the embryonic development of E. nani encompass egg-cleavage, egg-blastula, egg-gastrula, egg-nauplius, egg-zoea and egg-megalopa, and can be completed within 12 days. Unlike other freshwater crab, E. nani larvae hatched at megalopa stage and took a few hours to metamorphose to juveniles. The normal developmental stage is finally established for E. nani
