Analysis of cytotoxic activity of the CD4+ T lymphocytes generated by local immunotherapy

Abstract: Summary We previously reported that the anti-tumour effect of OK-432 is considerably enhanced by its intratumoral injection together with fibrinogen. In the present study, we generated killer T cells by culturing tumour-infiltrating lymphocytes from thyroid cancer patients who had received this local immunotherapy. Phenotypic analysis revealed that the T cells were positive for CD3+, CD4+, Leu8-, CD45RO+ and T-cell

“…mechanisms (Baker and Fosso, 1993). CD 45 RO positive lymphocyte levels, which are probably mostly memory T-lymphocytes (Justement, 1997;Katsumoto et al, 1996), decrease with increasing differentiation of thyroid carcinoma cells (Herrmann et al, 1994). In this study we studied differences in CD 45 receptor expression on the tumor infiltrating lymphocytes (TILs) as well as HLA class II expression in the various types of thyroid carcinomas and on TILs, with special reference to morphological parameters.…”

“…ICAM-1 cell surface expression is critical for lymphocyte adhesion to antigenpresenting cells (APCs) and for T cell-mediated cytolysis (1,3). Monoclonal antibody experiments have shown that the loss of ICAM-1 /LFA-1-mediated adhesion can prevent T cell adhesion and cytolysis of targets (6)(7)(8)(9). Cellular transfection experiments have demonstrated that interaction between ICAM-1 and the LFA-1 receptor is critical for effective HLA class II-restricted and allospecific T cell activation (10).…”

“…There is mounting evidence that ICAM-1 /LFA-1-mediated cell adhesion plays a critical role in the binding of T lymphocytes and may participate in immune activation. A series of studies have shown that monoclonal antibodies directed at ICAM-1 can inhibit cytotoxic T lymphocyte (CTL), natural killer (NK), and lymphokine activated killer (LAK) cell-mediated lysis of targets in vitro (6)(7)(8)(9). Furthermore, transfection of the ICAM-1 gene into major histocompatibility complex (MHC)-deficient fibroblasts enhances stimulation of T cells (10).…”

“…[28] Other possible mechanisms include models whereby Th1 cells promote antigenspecific tumor cell death consequent to their interaction with antigen-presenting cells that present tumor antigen in the vicinity of class I deficient, tumor antigen-expressing tumor cells. Examples of the latter are Fas-mediated, antigen-restricted cytolysis by CD4+ T lymphocytes [29,30] or stimulation of NK cells or macrophages due to antigen-specific secretion of IFN-γ such as by TAA-specific Th1 cells [21,23]. Of interest, others have also found that immunogenicity in the mouse prostate reconstitution model (from which RM-1 cells were derived) has been shown to be associated with resistance to CTL-mediated lysis [31].…”
