Analysis of Common Cold Virus (Human Rhinovirus Serotype 2) by Capillary Zone Electrophoresis:  The Problem of Peak Identification

Abstract: Different preparations of human rhinovirus serotype 2 (HRV2), a common cold virus, were analyzed by capillary zone electrophoresis (CZE) in untreated fused-silica capillaries using borate buffer (100 mmol/L, pH 8.3) and sodium dodecyl sulfate (10 mmol/L) as additive to prevent wall adsorption. The electropherograms showed one major peak at 205- and 254-nm detection wavelengths. The identity of the peak as originating from native virus was confirmed by several indirect methods. Heating to 56 degrees C is known … Show more

“…These highly ordered structures easily undergo conformational modifications during infection upon exposure to low pH or to elevated temperature. [31][32][33][34] Mann et al demonstrated a CZE method for the analysis of an intact recombinant adenovirus. 35 They used a capillary coated with polyvinyl alcohol, which eliminates the need for detergent additives in the running buffer.…”

“…Okun et al [31,[140][141][142][143][144][145] are pioneers in this area. Investigating by CE, the complexes between monoclonal antibodies and receptor fragments with human rinovirus serotype 2 (HRV2) were demonstrated.…”

“…Okun et al evaluated a different method for peak identification for human rhinovirus (HRV). The peaks were identified using various indirect methods involving (i) RNase treatment followed by enzymatic digestion and (ii) immunodepletion with monoclonal antibody binding (44). In a separate study, subviral particles of HRV were separated and identified using fast spectral scanning in conjunction with immunodepletion (46).…”
