Analysis of Biological Fluids by Direct Combination of Solid Phase Extraction and Desorption Electrospray Ionization Mass Spectrometry

Abstract: A novel, solid phase extraction (SPE)-based sample preparation method was developed for desorption electrospray ionization (DESI) mass spectrometry. Conventional SPE sample preparation was followed by a custom elution procedure. The eluate was evaporated from the closing frit of the cartridge using a gas jet. Thus the analyte was concentrated on the surface of the frit, which is ideal for DESI analysis. Application of the above SPE protocol allowed the concentration of the analyte content of up to 1 L liquid s… Show more

“…The basic obstacles included the indispensable sample preparation, and also the incompatibility of ionization methods with the environment or the sample itself. The development of "direct" or "ambient" ionization methods has lifted some of these constraints [1], however the sensitivity of these methods ranging from laser ablation/desorption to desorption electrospray is generally poor [2,3]. It has soon been recognized that various desorption ionization methods produce a significant amount of partially charged aerosol besides gas-phase molecular ions.…”

“…It should be noted that the increase in sensitivity up to six orders of magnitude was reported for DESI-MS when solid-phase extraction was used for drug monitoring in human serum [40]. As another option to increase the sensitivity, derivatization techniques were applied to DESI-MS analysis of saccharides in biological fluids [37].…”
