Analysis of Bacterial Membrane Proteins Produced During Mammalian Infection Using Hydrophobic Antigen Tissue Triton Extraction (HATTREX)

Abstract: Hydrophobic antigen tissue Triton extraction (HATTREX) provides a method to extract and identify hydrophobic bacterial membrane proteins from host‐infected tissues. The non‐ionic detergent Triton X‐114 is used to solubilize host‐infected tissues at 4°C. Subsequent phase partitioning of Triton X‐114 extracted material at 37°C results in a “detergent poor” aqueous phase and a “detergent rich” detergent phase. While soluble proteins partition to the aqueous phase, hydrophobic proteins, such as proteins of the out… Show more

“…Our lab has successfully applied Triton X‐114 to the extraction of P‐glycoprotein from cells . Notably, while Triton X‐114 has been employed often for the extraction of membrane proteins from cultured cells, its application to tissue samples has been reported only rarely . In this study, this detergent was used to extract membrane proteins from breast tissue samples.…”

“…The proteome of pathogenic leptospires, the causative agent of leptospirosis, expressed during urinary excretion from reservoir hosts of infection, is modulated to facilitate host evasion by diminution of antigen expression and increased expression of the virulence factor Loa22 (Nally et al , 2007 ; Monahan et al , 2008 ). Furthermore, proteomic technologies are compatible with novel extraction procedures to enrich for bacterial hydrophobic outer membrane proteins expressed during infection (Nally et al , 2007 ; Crother and Nally, 2008 ). Finally, the continued development of novel proteomic approaches such as Capillary Electrophoresis-Mass Spectrometry (CE-MS) have the capability to identify panels of peptides that can be used for disease diagnosis and for differential diagnosis of the causative bacteria of the infections of the mammary gland (Mansor et al , 2013 ; Albalat et al , 2014 ).…”
