Analysis of bacterial community profiles of endodontically infected primary teeth using pyrosequencing

Abstract: This study revealed a relationship between the oral microbiome and endodontic infection in primary teeth. Additionally, anaerobic bacteria such as Fusobacterium species were dominant in the teeth with apical abscesses.

“…It was the pathogen that could cause periapical infection [43][44][45], often found in deep dentin caries [30,46] and infected root canals [42]. Similarly, Fusobacterium was often detected in deep dentin caries [30,46] and infected root canals [44,45,47], It was a strict obligate anaerobe, with negative gram staining, normally living in oral cavity, upper digestive tract, intestinal tract, urogenital tract of human or animal and soil. It was most commonly seen in oral dental plaque.…”

“…Similarly, Fusobacterium is often detected in deep dentin caries [30,45] and infected root canals [43,44,46]. It is a strictly anaerobic Gram-negative bacillus living in the oral cavity and the upper digestive, intestine and urogenital tracts of humans or animals, as well as in the soil.…”

“…Only teeth from adult patients with carious lesions, necrotic pulps, and radiographic evidence of apical periradicular disease were involved in this study. We excluded patients who had systemic disease, cancer, diabetes, immunodeficiency disorder, and a history of using antibiotics or fluoride in the previous 3 months [ 19 , 20 ].…”

“…Afterwards, the samples were transferred to the laboratory for DNA extraction in frozen ice packs. A negative control (sample code 19) using sterile file and paper points which were not applied to the root canal was used in parallel to test for the existence of contaminants in the file and paper points [ 19 , 22 , 23 ].…”
