The high number of vitamins and minerals in Pokea meat encouraged us to determine the effect of consuming Pokea meat on Nitric Oxide Plasma levels in patients with hypertension and normotension. This study aimed to analyze Pokea meat consumption (Batissa violacea var. celebensis von Martens) on plasma oxide (NO) levels in hypertensive patients in Sampara, Konawe District. This research uses an observational analytic method with a case-control study design through molecular biology approach. The sample comprises 60 people consisting of 30 case samples and 30 control samples using the purposive sampling technique. Laboratory examination data is on NO plasma levels. Statistical analysis used data analysis use-dependent t-test. The distribution of Pokea meat consumption variables in the Hypertension group respondents had a mean value of 35.14 ± 17.66, while in the Non-Hypertension group of respondents was 41.10 ± 19.82. In the variable nitric oxide, the Hypertension group had a mean and standard deviation of 69.48 ± 42.78 µmol / L while the Non-Hypertension group had a mean and standard deviation of 262.8 ± 39.90 µmol / L. The statistical test analysis results showed an effect of Pokea consumption on plasma NO levels (p = 0,000). Pokea Consumption Influences NO Plasma Levels in Hypertension Patients, and there are also differences in NO Plasma Levels in Patients with Hypertension and non-hypertension in Sampara District, Konawe District, Southeast Sulawesi.

BACKGROUND: At present, the β-catenin gene is a genetic change considered as the most important factor in the initiation and the progress of cancer. Bivalve extract contains steroid compounds that are thought to have an aphrodisiac and anti-inflammatory effect. The triterpenoid component detected in the crude bivalves extract is thought to have antitumor activity. AIM: This study aimed at knowing the effect of ethanol (EtOH) extract, ethyl acetate fraction, and the n-hexane fraction of pokea clam on β-catenin gene mutation in mice induced by azoxymethane (AOM) and dextran sulfate sodium (DSS) using the polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method. METHODS: The testing of the effect of pokea clam extract was conducted on several treatment groups, namely, AOM/DSSinfected mice treated with EtOH extract, ethyl acetate fraction, and N-hexane fraction. The positive control and the negative control of the three treatment groups were done by giving the extract with a dosage of 0.25 mg/bb to mice for 14 days. RESULTS: Based on the result of β-catenin gene amplification using the PCR method and RFLP analysis, the EtOH extract and ethyl acetate fraction groups had the same band, namely, 56 bp and 12 bp, consecutively; meanwhile, the n-hexane fraction was marked by the band formation of 191 bp, 82 bp and 48 bp, 69 bp, 116 bp, 151 bp, 180 bp, and 234 bp. From the types of band and the band size that have been formed, it shows that they have the same formation and similarities in the positive and negative treatment characterized by the amplification that occurred in the band size of 227 bp, and the result of amplification from RFLP analysis showed the same band with the same band size, namely, 89 and 138 bp. CONCLUSION: Based on this, it can be concluded that N-hexane fraction affects β-catenin gene mutation at the first or second base at codon position 32 or 33, and the third base, namely, the bands of 82 and 145 bp that also characterize the effect of N-hexane on β-catenin gene mutation at the second or third base. It is supported by the sequencing method obtaining the homological percentage of 99.13%.

Batissaviolaceacelebensisis an endemic species of shellfish on Sulawesi Island. This animal is one of the staple foods of the people living around the Konaweha River. Various researches have been conducted to find the contamination of Cadmium, Plumbum and Hydragryrum heavy metals on the Konaweha River. Heavy metals can cause bioaccumulation in Batissaviolaceacelebensis. The objective of this study was to analyze the environmental health risks of heavy metals exposure in humans who consumed Batissaviolaceacelebensisfrom the Konaweha River. The study employed quantitative research (Field Research and Research Laboratory). By using the Environmental Health Risk Analysis method adopted from USEPA. The results showed that the concentration of heavy metals in Batissaviolaceacelebensis was 0.03345 mg/kg (cd), 0.04505 mg/kg (Pb) and 0.001833 mg/kg (Hg). The value of human intakes is 0.0054235 mg/kg/day (cd), 0.0073076 mg/kg/day (Pb) and 0,0002962 mg/kg/day (Hg). The duration of exposure for 30 years, the average intake rate of 68.09 grams/day, the average exposure frequency is 124 days/year. So that the Risk Quotient value <1 is obtained. Pollution in the Konaweha River has contaminated Batissavoilecaecelebensis. It is predicted that the risk of contamination will be felt by humans if they consume more than 30 years. It would be better if the Risk Quotient value was lowered by minimizing the intensity of exposure to consumption of Batissaviolaceacelebensis.
