Anakinra Reduces Epileptogenesis, Provides Neuroprotection, and Attenuates Behavioral Impairments in Rats in the Lithium–Pilocarpine Model of Epilepsy

Abstract: Temporal lobe epilepsy is a widespread chronic disorder that manifests as spontaneous seizures and is often characterized by refractoriness to drug treatment. Temporal lobe epilepsy can be caused by a primary brain injury; therefore, the prevention of epileptogenesis after a primary event is considered one of the best treatment options. However, a preventive treatment for epilepsy still does not exist. Neuroinflammation is directly involved in epileptogenesis and neurodegeneration, leading to the epileptic con… Show more

“…We hypothesized that the use of MTEP could affect the expression of astroglial proteins and different subunits of NMDA- and AMPA-receptors at the latent and chronic phases of the model. The production of GFAP was increased in the hippocampus of the Pilo rats in line with previous studies in the lithium–pilocarpine model [ 34 , 57 ]. However, the MTEP treatment had almost no effect on this change.…”

“…Neurodegeneration in the dorsal hippocampus is often observed in the lithium–pilocarpine model [ 33 , 34 ]. MTEP demonstrated a significant neuroprotective effect in a kainate seizure model [ 24 ].…”

“…The lithium–pilocarpine model of temporal lobe epilepsy was used in this study. The effectiveness of the used model protocol was demonstrated in our earlier research [ 52 ]. One day before pilocarpine injection, rats were injected with lithium chloride (intraperitoneally (i.p.…”

“…Interestingly, transient treatment with anakinra after brain injury has been reported to provide disease-modifying or antiepileptogenic effects in non-infectious post-status epilepticus models of acquired epilepsy ( Noé et al, 2013 ; Dyomina et al, 2020 ). Similarly, the immunomodulator fingolimod, which is established as an MS therapy, was reported to exert antiepileptogenic, neuroprotective effects, and anti-inflammatory effects in post-status epilepticus models of TLE ( Gao et al, 2012 ; Pitsch et al, 2019 ).…”
