Anabolism versus catabolism of [5-3H]uridine and its relationship to ribonucleic acid labelling in mouse liver after partial hepatectomy

Abstract: The balance between anabolism and catabolism of [5-3H]uridine was studied in the mouse after partial hepatectomy. Labelling of RNA and UDP-glucose was determined and evaluated in relation to changes in the specific radioactivity of UTP. The amounts of labelled catabolic products of uridine were increased several-fold in liver and blood after partial hepatectomy. The specific radioactivity of RNA decreased to about 60% of the control value at 6h and was in the same range as that of control liver at 24h after op… Show more

“…The radioactivity of UTP was determined in the neutralized acid-soluble fraction after homogenization in 0.4 M HCIO... The amount of UTP used for calculation of the UTP-specific radioactivity was 257 nmol/g wet weight (33). Specific radioactivity of RNA was determined in the precipitated acid-insoluble material obtained after homogeniza tion of the liver tissue in the 0. by others (5).…”

“…In contrast with regenerating rat liver, mouse liver reveals no enhanced uptake of orotic acid or incorporation of this precursor into either UTP or RNA at 6 and 24h after partial hepatectomy (Yngner et al, 1979a(Yngner et al, , 1980, nor is there an increased incorporation of labelled uridine into UTP or RNA of regenerating mouse liver, despite an increased uptake of the precursor into the total acid-soluble fraction (Yngner et al, 1979b).…”
