Hydrazinonicotinamide (HYNIC) forms stable coordination complexes with Tc-99m when reacted with Tc(V)oxo species such as Tc-mannitol or other Tc-polyhydric complexes. However, radio-HPLC of [Tc-For-MLFK-HYNIC] labeled via Tc-polyhydric ligands demonstrated multiple radiochemical species each with unique biodistribution patterns. This is likely due to the fact that Tc can bind to the hydrazino moiety, as well as polyhydric ligands, in a variety of coordination geometries. Tridentate ligands, such as bis(mercaptoethyl)methylamine (NS2), may constrain the possible coordination geometries and improve overall stability. To investigate this, we synthesized NS2, converted the [Tc-mannitol-For-MLFK-HYNIC] to the corresponding NS2-containing complex [Tc-NS2-For-MLFK-HYNIC], and compared its infection imaging and biodistribution properties with [Tc-mannitol-For-MLFK-HYNIC]. Conversion to the NS2 complex was confirmed by HPLC which showed a single unique hydrophobic species with retention time greater than the [Tc-mannitol-For-MLFK-HYNIC] complex. Imaging experiments with both preparations were performed in rabbits with E. coli infections in the left thigh. Tissue radioactivity measurements demonstrated that compared to Tc-mannitol-peptide, accumulation of Tc-NS2-peptide was lower in blood, heart, and normal muscle and higher in spleen, infected muscle, and pus (p < 0.01). These results indicate that the Tc-NS2-peptide complex is chemically more homogeneous and exhibits improved infection localization and biodistribution properties. In an effort to model the interactions of the metal-HYNIC core with NS2 and related ligand types, the reactions of [ReCl3(NNC5H4NH)(NHNC5H4N)] and [99TcCl3(NNC5H4NH)(NHNC5H4N)], effective structural analogues for the [M(NNC5H4NH(x))2] core, with NS2, C5H3N-2,6-(CH2SH)2, O(CH2CH2SH)2, and S(CH2CH2SH)2 were investigated and the compounds [M[CH3N(CH2CH2S)2](NNC5H4N)(NHNC5H4N] (M = 99Tc (5a), Re (5b)), [Re[C5H3N-2,6-(CH2S)2](NNC5H4N)(NHNC5H4N)].CH2Cl2.0.5MeOH (7), [Re[SCH2CH2)2O] (NNC5H4N)(NHNC5H4N)] (8), and [Re[(SCH2CH2)2S](NNC5H4NH)(NHNC5H4N)]Cl (9) were isolated. Four novel rhenium complexes of formula [ReCl(4)(bpym)] (1), [ReBr(4)(bpym)] (2) PPh(4)[ReCl(4)(bpym)] (3) and NBu(4)[ReBr(4)(bpym)] (4) (bpym = 2,2'-bipyrimidine, PPh(4) = tetraphenylphosphonium cation and NBu(4) = tetrabutylammonium cation), have been synthesized and their crystal structures determined by single-crystal X-ray diffraction. The structures of 1 and 2 consist of [ReX(4)(bpym)] molecules held together by van der Waals forces. In both complexes the Re(iv) central atom is surrounded by four halide anions and two nitrogen atoms of a bpym bidentate ligand in a distorted octahedral environment. The structures of 3 and 4 consist of [ReX(4)(bpym)](-) anions and PPh(4)(+) () or NBu(4)(+) (4) cations. The coordination sphere of the Re(iii) metal ion is the same as in 1 and 2, respectively. However, whereas the Re-X bonds are longer the Re-N bonds are shorter than in 1 and 2. This fact reveals that the bpym ligand forms a stronger bond with Re(iii) than with Re(iv) resulting in a stabilisation of the lower oxidation state. [ReX(4)(bpym)] complexes are easily reduced, chemically and electrochemically, to the corresponding [ReX(4)(bpym)](-) anions. A voltammetric study shows that the electron transference is a reversible process characterized by formal redox potentials of +0.19 V (1) and +0.32 V (2) vs. NHE, in acetonitrile as solvent.

The new compound {(PhS)(2)ReO(mu-SPh)}(2), 1, was synthesized from Re(2)O(7) and PhSH and then used as the synthon for a number of hitherto unknown oxorhenium(V) compounds. Reactions between dithiols and 1 (2:1 ratio) afford {PhSReO(dt)}(2), where the dithiols, dtH(2), are 1,2-ethanedithiol (edtH(2)), 1,3-propanedithiol (pdtH(2)), 1,3-butanedithiol (pdtMeH(2)), 1,2-benzenedithiol (bdtH(2)), 2-(mercaptomethyl)thiophenol (mtpH(2)), and 2-mercaptoethyl sulfide (mesH(2)). Similar reactions carried out with a 3:1 ratio of dtH(2) to 1 afford [(ReO)(2)(dt)(3)], dt = edt, pdt. When NEt(3) was introduced prior to the 3:1 reaction between edtH(2) and 1, a compound containing an anionic complex was isolated, [PPh(4)][ReO(edt)(2)]. The new compounds were characterized analytically, spectroscopically, and crystallographically. The Re-O groups in two of the compounds, 1 and {ReO(mu-SPh)(bdt)}(2), exist in rare anti orientations; the others adopt the more familiar syn geometry, as discussed. Selected monomerization reactions of {PhSReO(dt)}(2) were also carried out: {PhSReO(dt)}(2) + 2L = 2[PhSReO(dt)L]. The rate for L = 4-phenylpyridine is given by v = {k(a)[L] + k(b)[L](2)} x [{PhSReO(dt)}(2)], as it is for the reactions of {MeReO(dt)}(2); for all of these compounds, the reaction proceeds nearly entirely by the third-order pathway. Values of k(b)/L(2) mol(-2) s(-1) at 25.0 degrees C are 5.8 x 10(2) (mtp), 2.97 x 10(3) (pdt), 4.62 x 10(5) (edt), and 3.87 x 10(5) (bdt). The rate law for the reactions of {PhSReO(dt)}(2) with L = PAr(3) is v = k(a)[L]/{1 + kappa[L]} x [{PhSReO(dt)}(2)]. For PPh(3), values at 25.0 degrees C of k(a)/L mol(-1) s(-1) (kappa/L mol(-1)) for {PhSReO(dt)}(2) are 9.64 x 10(-2) (1.87) for mtp, 3.43 x 10(-2) (0.492) for pdt, 1.91 (1.42) for edt, 1.84 x 10(-2) (0.82) for bdt, and 1.14 x 10(3) (10.6) for 1. Mechanisms are proposed that are consistent with the data obtained and with earlier work.
