An Overview on the Toxic Morphological Changes in the Retinal Pigment Epithelium after Systemic Compound Administration

Abstract: Many medications that are administered systemically for nonocular conditions may evoke ocular toxicological complications. Therefore, the eye is routinely investigated histopathologically in preclinical in vivo toxicity studies. The retinal pigment epithelium is a likely target for systemically administered compounds, since the underlying choroid is highly vascularized. The specialized pigment epithelium has numerous functions that all maintain the integrity and function of photoreceptors. Consequently, toxic … Show more

“…The RPE layer mainly serves as a barrier between the retina and the circulatory system 25. An increase in the thickness of this layer is well documented in age-related retinal degeneration studies, in which retinal pathology is a result of chronic inflammatory damage 26 27.…”

“…The observed symptoms include night blindness, photopsia, blurred vision, and visual impairment, which are likely associated with retinal dysfunction (20,21). Two independent reports of rodent models suggest that HSP90 plays a critical role in retinal function and that sustained HSP90 inhibition might adversely affect visual function (34,49). Therefore, ocular effects may be minimized by reducing retinal exposure.…”
