Sheeppox virus (SPPV) is one of the listed and notifiable disease affect sheep production with major effect on the trade of new breed of sheep. This study was conducted to identify sheep pox by using cell culture, electron microscope (EM) and open reading frame (ORF) 103 gene during an outbreak of local breed of sheep occurred in Sharkia Governorate, Egypt in April 2017. Affected adult sheep showed typical skin pox lesion on face, the inner side of lips, inner aspect of the thigh and under the tail. The incidence rate of infection was 23.5% and the mortality rate in young lambs aged 3 to 6 months old was 8.2%. Forty-three scabs and tissue samples from clinically diseased adult sheep and dead lambs were collected and subjected to culture on Vero cell. The cytopathic effect (CPE) was observed within 3 to 7 days in 40 samples. A typical Poxvirus was a brick-like shape with round ends by negative staining of EM and ovoid like structure with dumb-bell shaped DNA core with concave bodies sides by positive staining of EM. By conventional PCR utilizing ORF103 gene and obtained bands of about 570 bp referred to SPPV. The sequence amplicon was analyzed by NCBI-BLAST and register in GeneBank under accession N. MG873537 and phylogentic tree was designed which revealed that the isolated strain of SPPV was resembled with other strains of SPPV isolated in Egypt, India, China, and USA. Finally, both EM and PCR are considered as sensitives, rapids, and powerful methods to identify SPPV from tissue and scab's samples without the need of further culture in addition to the useful and easily use of ORF103 gene to differentiate SPPV from other Capripoxvirus.

Aim:To isolate, identify, and differentiate Capripoxviruses (CaPV) (sheep pox virus and goat pox virus) infections by egg inoculation, transmission electron microscopy (TEM), and 30 kDa RNA polymerase subunit gene-based polymerase chain reaction (PCR) (RPO30) in clinically affected animals in Hawamdia township of Giza Governorate, Egypt.Materials and Methods:A total of 37 scab samples were collected from clinically suspected field cases of sheep pox and goat pox. These samples were collected during (2014-2015) during different outbreaks of sheep pox and goat pox from Hawamdia township of Giza Governorate, Egypt. The samples were subjected to egg inoculation, TEM, and (RPO30) gene-based PCR. By using the egg inoculation: Previously prepared 37 scab samples (n=23 sheep and n=14 goats) were inoculated on the chorioallantoic membrane of specific pathogen free (SPF) embryonated chicken eggs (12 days old age). In the presence of the suitable percentage of humidity and candling, the inoculated eggs were incubated at 37°C. By using the TEM: Samples showed positive pock lesions on the chorioallantoic membranes, were fixed in glutaraldehyde, then processed and sectioned for TEM. Using the (RPO30) gene-based PCR assay, 30 of positive samples after egg inoculation (n=19 sheep and n=11 goats) were screened.Results:Using the egg inoculation, a characteristic pock lesions for poxviruses were seen in 30/37 (n=19 sheep and n=11 goats) (81.08%). Using the TEM, examination of the positive samples after egg inoculation revealed positive result in 23/30 (n=15 sheep and n=8 goats) (76.66%). The positive results represented by the presence of negatively stained oval-shape virus particles. Using the (RPO30) gene-based PCR assay, out of 30 total of positive samples after egg inoculation (n=19 sheep and n=11 goats) were screened, 27 (90%) samples (n=17 sheep and n=10 goats) were positive. The given band sizes of sheep and goats were 172 and 152 bp, respectively.Conclusion:PCR assay depended on RPO30 gene can be used lonely for the detection, identification, and differentiation of CaPVs. RPO30 gene-based PCR assay in combination with gene sequencing helps in molecular epidemiological studies of CaPV infection.

The sheeppox virus belongs to the Capripoxvirus genus, which is one of the eleven genera of the subfamily Chordopoxviruses of the family Poxviridae. The genus Capripoxvirus consists of the sheeppox virus (SPPV), goatpox virus (GTPV), and lumpy skin disease virus (LSDV), which are sources of dermatological diseases in sheep, goats, and cattle, respectively [3].The viral genome is a double-stranded linear DNA of 150 kbp [4]. Each of the three species of the genus Capripoxvirus infects their natural hosts and is named according to the host origin; nevertheless, cross-species infections among sheep, goats, and cattle have been detected [5,6]. This makes differential diagnosis more difficult, and serological diagnosis based on detection of antibodies by immunofluorescence, ELISA, or gel immunodiffusion is inefficient [7]. In addition, serological methods cannot distinguish sheeppox infection from lumpy skin or goatpox disease [8]. Only molecular methods allow distinction between all these species through sequencing and molecular analysis of some viral genes [9,10].Several trials have been carried out to establish the relationship among these virus species by constructing phylogenetic trees after sequencing RPO30, P32, ORF117, and other genes [5,[11][12][13]. None of these genes have given the consistent results required to create a reference candidate to anchor phylogenetic classification within the genus Capripoxvirus. Therefore, it is essential to find a common gene that allows more relevant phylogenetic classification and distinction between these virus species without producing discrepancies between studies. Materials and methods Sample collectionSkin specimens with crusted scab lesions from the axilla and/or tail were collected from sheep suspected to be infected with pox virus (Table 1). Eleven samples were received from different regions of Tunisia and collected between 2010 and 2016, a period in which more than 344 outbreaks were reported [2].
