Colibacillosis is considered important in the poultry industry because it generates economic losses due to the disturbance of growth, the decline in production, an increase number of culled chicken, and reduced quality of carcasses and eggs. Study of colibacillosis and antibiotics resistance patterns in broiler was conducted at the Indonesian Research Centre for Veterinary Science. The aim of the study was to provide the latest information on the status of the presence of Escherichia coli in broiler farms in four districts in West Java Province as well as its antibiotics drug resistance patterns and their pathogenicity will be studied. A total of 196 samples of the intestine, liver, heart, egg yolk, air sac that showed gross abnormalities were sampled and feces were collected and used in this study. Escherichia coli was isolated for characterisation and antibiotics drug resistance patterns and their pathogenicity were investigated. Escherichia coli were recovered from 149 (76.02%) samples out of the total samples collected. In this present study, one (7.7%) E. coli isolate was serologically typed into O 78 and 12 of them were untypable whereasthe other 7 (4.93%) isolates were serologycally typed into O 157 and 135 were untypable. In vitro pathogenicity indicated that 91.55% of the isolates were positive for Congo red binding assay and 4.93% isolates were positive for haemolysine production. Antibiogram profiles indicated that 98.70, 79.30, 75.10, 61.40, and 57.70% isolates were resistance against to amphicillin, neomycin, streptomycin, sulfamethoksazol trimethropim, and kanamycin, respectively. A total of 149 (76,02%) histopathologically had consistent colibacillosis lesion with various degree of severity.

The aim of the present study was to investigate the genetic diversity and antimicrobial resistance (AMR) of E. coli during enrofloxacin therapy in broilers affected by colisepticemia. Three unrelated farms with ongoing colibacillosis outbreaks were sampled at day 1 before treatment and at days 5, 10 and 24 post-treatment. A total of 179 E. coli isolates were collected from extraintestinal organs and submitted to serotyping, PFGE and the minimum inhibitory concentration (MIC) against enrofloxacin. PFGE clusters shifted from 3–6 at D1 to 10–16 at D5, D10 and D24, suggesting an increased population diversity after the treatment. The majority of strains belonged to NT or O78 and to ST117 or ST23. PFGE results were confirmed with SNP calling: no persistent isolates were identified. An increase in resistance to fluoroquinolones in E. coli isolates was observed along the treatment. Resistome analyses revealed qnrB19 and qnrS1 genes along with mutations in the gyrA, parC and parE genes. Interestingly, despite a fluoroquinolone selective pressure, qnr-carrying plasmids did not persist. On the contrary, two conjugative AMR plasmid clusters (AB233 and AA474) harboring AMR genes other than qnr were persistent since they were identified in both D1 and D10 genomes in two farms. Further studies should be performed in order to confirm plasmid persistence not associated (in vivo) to antimicrobial selective pressure.

Abstract:Salmonella species (spp) and Escherichia coli (E. coli) are the most common infectious pathogens in poultry. Antimicrobials were given either for the treatment or growth promoters that can increase the possibility of emergence of bacterial resistance towards antimicrobials. The aim of this study was to determine the prevalence of antimicrobial resistant (AMR) Salmonella spp and E. coli isolated from a sample of broiler farms in East Coast Malaysia from 2018-2019. A total of 384 cloacal swabs were collected from broilers farms in Kelantan, Terengganu, and Pahang. The bacteria were isolated and confirmed by bacteriological and serological methods. Following that, confirmed isolates were subjected to antimicrobial susceptibility test. Salmonella spp and E. coli were recovered from the cloacal swabs samples with the overall prevalence of 6.5% and 51.8% respectively. In Kelantan, Terengganu and Pahang, the prevalence of Salmonella spp were 7%, 6.5% and 5.8% respectively, while the prevalence for E. coli were 50%, 48.3% and 58% respectively. Salmonella spp and E. coli displayed resistance towards the following antimicrobials: erythromycin (100% for both pathogens), chloramphenicol (76.2%, 84.5%), tetracycline (62%, 94.6%), ampicillin (47.7%, 87%), sulfamethoxazole/trimethoprim (42.9%, 83.3%), ciprofloxacin (4.8%, 23.8%), nalidixic acid (9.6%, 60.7%), streptomycin (19%,66%), and kanamycin (28.6%,57%), cephalotin (0%, 11%), gentamicin (0%, 20.2%) respectively. No resistance were recorded towards colistin for both pathogens. Multidrug resistance (MDR) was recorded in 82% of Salmonella spp and 100% of E. coli. These findings demonstrate the high prevalence of MDR Salmonella spp. and E. coli in broiler farms in East coast Malaysia. This could be attributed to the excessive use of antimicrobial agents by the poultry farm owners. Enhanced control measures and a strong monitoring system should be urgently implemented to reduce the emergence of antimicrobial resistance that is harmful to public health.
