An oralSalmonellavaccine promotes the down-regulation of cell surface Toll-like receptor 4 (TLR4) and TLR2 expression in mice

Abstract: A single oral immunization with the Lon-protease-deficient Salmonella enterica serovar Typhimurium (strain CS2022) induced protective immunity in mice against a subcutaneous challenge with virulent Listeria monocytogenes as well as virulent Salmonella serovar Typhimurium. The populations of cell surface Toll-like receptor 4 (TLR4) and TLR2 on peritoneal macrophages decreased at week 6 after immunization. This population decrease was not reversed after a challenge with either Salmonella or Listeria. These resul… Show more

“…Thus, 7-week-old mice were s.c. infected with a total of 1 ϫ 10 7 CFU of stationary-growth-phase GAS into both hind footpads (5 ϫ 10 6 CFU per footpad). After s.c. infection, the survival rates were observed every 24 h for 336 h postinfection, or the number of bacteria in the blood and tissue samples including the popliteal lymph nodes (PLN), spleen, and liver were determined by plating onto sheep blood agar (11,27). All mice were bred at the animal facility at the Kitasato Institute, and all mouse experiments were performed in accordance with institutional protocol guidelines under an approved protocol.…”

“…As bacterial Lon has been shown to be important for the infectivity and/or pathogenicity of certain species such as B. abortus and S. typhimurium, compounds that specifically block bacterial but not human Lon, hold promise for a new class of anti-microbial agents (8,31,33,34,99,102). In addition, mitochondrial Lon has been implicated in the response to oxidative stress and damage to proteins and mtDNA (7,46,103), which has direct relevance to aging.…”

“…TLR4 is often overexpressed in malignant and tumor-infiltrating immune cells, and the application of TLR4 ligands in cancer therapies is desirable for enhancement of antitumor immunity [38, 39]. PAX5 haploinsufficiency induced decreased expression of TLR4, and the downregulated TLR4 participated in immune tolerance [43, 44]. Fos was involved in inducing a large number of cytokine genes and other genes that were central to the productive immune response.…”
