An Optimized Method to Generate Human Active Osteoclasts From Peripheral Blood Monocytes

Abstract: Osteoclasts (OCs), the bone-resorbing cells, play a key role in skeletal development and adult bone remodeling. They also participate in the pathogenesis of various bone disorders. One of the major technical difficulties in the generation of OCs, when working on human material, is the ability to achieve large differentiation of mature OCs from human peripheral blood mononuclear cells (PBMCs). Access to a standardized source of active OCs is needed to better analyze the roles of human OCs. The aim of this study… Show more

“…6G). Because RANKL has the ability to stimulate OC formation (22,23), we hypothesized that the supernatants of RA FLS stimulated with cNETs would promote OC formation. To test this hypothesis, PB CD14 + monocytes isolated from healthy volunteers were incubated in the presence or absence of supernatants of FLS stimulated with either NETs or cNETs.…”

“…In in-vitro culture, under the influence of M-CSF and RANKL treatment, PBMCs differentiate to macrophage/pre-OCs, which is followed by cell fusion. Mature and functional OCs are characterized by the presence of multiple nuclei in a very large unified cell (multi-nucleated giant cells) which produce TRAP (TRAP stain positive) (Fig 2A and 2B) [38][39][40][41][42].…”

“…Osteoclasts are bone‐resorbing cells that positively function in skeletal‐related issues as well as the remodeling of adult bone, and they have a characteristic ability to reuptake bone tissues . A former study found that miR‐155‐deficient mice showed reduced bone destruction, attributed to reduced generation of osteoclasts .…”
