An L1-like molecule, the 8D9 antigen, is a potent substrate for neurite extension.

Abstract: The 8D9 antigen, a cell surface protein isolated from chicken brain that is related to the Li class of cell adhesion molecules, is shown to contain an activity that promotes the attachment of neurons and the outgrowth of neurites from chicken tecta and mouse cerebellum. When purified 8D9 antigen is attached to a nitrocellulose-coated substrate, neurons rapidly attach and extend unfasciculated neurites. Little or no attachment of astroglia, oligodendroglia, and fibroblast-like cells to the 8D9 antigen is observ… Show more

“…Production and characterization ofthe N-CAM antibodies have been described previously (Lagenaur and Lemmon, 1987;Chung et al, 199 I). The monoclonal antibody 12F8 recognizes polysialic acid epitopes on N-CAM in mammalian as well as avian species, and identifies embryonic forms of N-CAM distinct from adult forms lacking polysialic acid (Lagenaur et al, 1988).…”

“…Binding of Collagen XVIII to Retinal Basement Membranes-A PAPpen (EMS, Washington, PA) was used to circle an ϳ1-cm 2 area of a plastic dish coated with nitrocellulose (20). The hydrophobic ring reduces the volume of the incubation solutions to less than 100 l. Retinal basement membranes were isolated from E6 chick retinae as flat sheets within the encircled areas of the dishes as described (25).…”

“…Among the many cues that guide the axons of embryonic neurons to their proper destinations, axon fasciculation is an important developmental mechanism. The cell adhesion molecules (CAMs) involved in neurite fasciculation 43,44 include neural cell adhesion molecule (N-CAM) and NGF-inducible large external glycoprotein in the rat, neuron-glial cell adhesion molecule, G4, and 8D9 in the chicken, and L1 in the mouse. The role of CAMs on a given axon is to promote the extension of other neurites along this axon that, in turn, produces fasciculated axons.…”
