An iridium oxide nanoparticle and polythionine thin film based platform for sensitive Leishmania DNA detection

Abstract: An impedimetric label-free genosensor for high sensitive DNA detection is developed. This system is based on a screen-printed carbon electrode modified with the thionine layer and iridium oxide nanoparticles (IrO 2 NP). An aminated oligonucleotide probe is immobilized on the IrO 2 NP/polythionine modified electrode and ethanolamine was used as a blocking agent. Different diluted PCR amplified DNA samples have been detected. The selectivity and reproducibility of this system are studied and the system was highl… Show more

“…3B). Iridium oxide nanoparticles (IrO2NPs) are other 0D nanomaterials used due to their catalytic properties towards water oxidation reaction 26 , the production of oxygen from water, and their application in impedimetric sensors [27][28] . Impedance is a technique used to measure frequency changes on the dielectric medium close to the nanoparticles.…”

“…Earlier publications addressed detection of nucleic acid of infectious agents using voltammetric, amperometric and impedance electrochemical approaches (Hassen et al, 2008;Pournaghu-Azar et al, 2008;Cash et al, 2009;Benvidi et al, 2015). Notably, DNA hybridization biosensors based on electrochemical spectroscopy impedance (EIS) are being developed in both label-free formats and label-based approaches using mostly gold nanoparticle (AuNP) and cadmium sulfide quantum dot (CdS QD) as tags (Wang et al, 2002 and2003;Xu et al, 2004;Pumera et al, 2005;Bonanni et al, 2008;Rasheed and Sandhyarani, 2017 ;Ribovski et al, 2017) also employing other nanomaterials (Wang et al, 2003;De la Escosura-Muñiz and Merkoçi, 2010;Merkoçi, 2010;De la Escosura-Muñiz and Merkoçi, 2014;Mayorga-Martinez et al, 2015;De la Escosura-Muñiz et al, 2016). “…Therefore, these electrodes are cumbersome to use for electrochemical aptasensing applications. The screen printed carbon electrode (SPCE) based biosensor offers the advantages over the conventional electrochemical biosensors in terms of disposability, portability and feasibility for on-site analysis at remote areas [14][15][16]. The preparation of SPCEs is simple, inexpensive and versatile with advantages of mass production and miniaturization of devices.…”
