The objective of the present work was to obtain and evaluate the solid inclusion complexes of a-, b-cyclodextrins and hydroxypropyl-a-, hydroxypropyl-b-cyclodextrins derivatives with uracil (U) and anti-cancer agent 5-fluorouracil (5FU). The thermogravimetry (TG), differential scanning calorimetry (DSC), Fourier transform infrared spectroscopy in attenuated total reflectance mode (FTIR-ATR) and scanning electron microscopy (SEM) were used to characterize these inclusion compounds. New information on the physicochemical behavior of these solid state complexes synthesized by melting in solution was obtained. Corelating the experimental data we found that the complexation occurs for each host:guest system thus, the preparation method exhibited a real efficiency to interaction achievements. The inclusion capability and the thermal stability of the studied complexes were due to both the flexibility and the size of the cyclodextrin ring as well the stereoelectronic effect of the U or 5FU molecule. We appreciated that the complexes between U or 5FU with 2-hydroxypropyl derivatives of a- and b- CD have the largest content of guest and are the most stable. The obtained results could be useful in formulation of the pharmaceutical products and also in drug delivery.

Puerarin, which was used as a natural isoflavone in this study, was obtained from the Kudzu plant. The inhibitory effect of puerarin on acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) enzymes was investigated. The IC50 values of AChE (177.73 nM) and BChE (315.07 nM) were determined. The binding mechanism of puerarin, which has a high inhibitory effect on the active site of the enzyme, was demonstrated by the molecular docking method. The antioxidant activity was evaluated for the same molecule. For this purpose, Fe3+‐Fe2+ reducing capacity, ABTS radical scavenging activity, CUPRAC method, DPPH⋅ scavenging activity and DMPD radical scavenging method were used. The percentage (%) inhibition amount of puerarin molecule was calculated as 99.33 % for ABTS radical scavenging activity, which is the more preferred method for natural products. The absorbtion value of iron reduction of Puerarin was 0.122±0.010, and the value of copper reduction was 0.633±0.026. Our research efforts include uncovering the potent antioxidant potential of puerarin and, simultaneously, a comparative study of tacrine, primarily emphasizing its inhibitory effects against AChE and BChE enzymes. The puerarin is expected to be a guide for drug discovery against many problems, especially oxidative cell damage in Alzheimer's disease, mitochondrial dysfunction (MD), cancer, and cardiovascular diseases.

Puerarin is the major bioactive ingredient isolated from the dry root of Pueraria lobata , a plant used in traditional Chinese medicine. Puerarin has been used to treat diabetes and cataracts in China; however, its underlying mechanism of action remains unclear. The aim of the present study was to investigate the effectiveness and mechanism of puerarin in preventing cataracts in diabetic rats. Diabetes was induced by streptozocin (STZ) administration and rats were intraperitoneally injected with puerarin (25, 50 and 100 mg/kg). Blood glucose levels and cataract development were examined in the different experimental groups. In addition, the expression levels of markers associated with oxidative stress, including nuclear factor erythroid 2 like 2 (Nrf2) and heme oxygenase-1 (HO-1), were analyzed. The present results suggested that treatment with puerarin at 25, 50 and 100 mg/kg significantly reduced blood glucose levels and the incidence of cataract in STZ-induced diabetic rats. Additionally, puerarin treatment reduced oxidative stress, restoring the levels of malondialdehyde and glutathione, and the activity of glutathione peroxidase. Furthermore, puerarin administration decreased the expression levels of retinal vascular endothelial growth factor and interleukin-1β and increased the mRNA expression levels of Nrf2 and HO-1, thus inhibiting oxidative stress. The present findings suggested that puerarin had hypoglycemic effects and that it prevented cataract development and progression in diabetic rats by reducing oxidative stress through the Nrf2/HO-1 signaling pathway.
