An integrated view of cisplatin-induced nephrotoxicity and ototoxicity

Abstract: Cisplatin is one of the most widely-used drugs to treat cancers. However, its nephrotoxic and ototoxic side-effects remain major clinical limitations. Recent studies have improved our understanding of the molecular mechanisms of cisplatin-induced nephrotoxicity and ototoxicity. While cisplatin binding to DNA is the major cytotoxic mechanism in proliferating (cancer) cells, nephrotoxicity and ototoxicity appear to result from toxic levels of reactive oxygen species and protein dysregulation within various cellu… Show more

“…Although cancer prevention is crucial, improving existing therapies and reducing harmful side effects is just as vital. Cisplatin is widely used to treat various solid malignancies; however, its use is limited due to its associated nephrotoxicity, which primarily occurs in kidney proximal tubule epithelial cells (Karasawa and Steyger, 2015). PCA was found to prevent induced oxidative damage in kidney proximal tubule rats could be due to the antioxidant activity of PCA, given that it has been postulated that oxidation plays a role in vascular impairment in diabetes.…”

“…This platinum complex causes the crosslinking of two DNA strands in cells, which prevents cell division and finally leads to programmed cell death. In addition to nephrotoxicity, neurotoxicity and ototoxicity are dose-limiting adverse effects of cisplatin treatment [74,75].…”

“…The antitumour and toxic effects of the drug are frequently discussed 1. Nephrotoxicity is the most common side effect of the drug's therapeutic effectiveness and is associated with high mortality 2. However, the mechanism of cisplatin‐induced AKI remains unclear.…”
