An Integrated Decoupler for Capillary Electrophoresis with Electrochemical Detection:  Application to Analysis of Brain Microdialysate

Abstract: An approach to capillary electrophoresis with electrochemical detection (CE-EC) suitable for determination of dopamine in 1-min brain microdialysate samples is described. The CE-EC system includes an electrochemical detection cell that permits easy, precise, and permanent alignment of a carbon fiber microelectrode with a separation capillary (30-micron i.d., 75-cm length). Amperometric detection was performed at a constant applied potential of 600 mV with respect to a Ag/AgCl reference electrode. Decoupling of… Show more

“…With the same technique it is possible to resolve the monoamine serotonin from DA [115]. DA was detected in brain microdialysates by CE-EC with low-mass (attomol) detection limit, and low-concentration (nM) detection limits [132]. The CA have been analyzed with CE-EC even in single cells.…”

“…Whereas CE with EC detection is a powerful technique, its performance is highly dependent on the design of the detector [6][7][8][9]. Alignment between the capillary bore and the electrode must be precise and easily maintained to ensure high sensitivity.…”

“…Interference between the separation potential and EC detection must also be limited to minimize noise and instability in the detector [2,7]. Several variations of EC detection for CE have been investigated; in particular, two main methods of isolating the electrode from the separation potential have been developed, off-column and end-column EC detection [2,7]. Off-column EC detection for CE utilizes a decoupling joint to reduce the interference of the high voltage electric field used in CE from the EC detection circuit.…”
