1,8‐Diazabicyclo[5.4.0]undec‐7‐ene immobilized on silica (SB‐DBU)Cl as a reusable and heterogeneous catalyst was applied for the one‐pot, three‐component synthesis of 2‐amino‐4,8‐dihydropyrano[3,2‐b]pyrane‐3‐carbonitriles and pyridopyrimidines, under solvent‐free conditions. The reaction was carried out at room temperature, and pure products were obtained in high yields and short reaction times (3–7 min). This work introduced an environmentally benign procedure for the synthesis of these classes of biological active compounds.

The main purpose of this review is to provide an overview of the current knowledge for development of new molecules of pyridopyrimidines by using different multi-component reactions that lead to give effective anticancer agents. Thus, the developed compounds have been derived from pyridine and pyrimidines fusion or from possible isomeric forms of pyridopyrimidine derivatives. They are obtained from different approaches of multistep synthesis reaction such as Tandemaza-witting reaction, annulations, and Biginelli type reactions. Apart from the synthetic schemes, their biological applications with an alternative mechanism of action, such as growth inhibitors by cell cycle arrest, apoptosis, inhibition of angiogenesis, disruption of cell migration, and mutation, are also understood. These receptors and enzymes are responsible for causes of cancer because disruptions of dihydrofolate reductase, tyrosine kinase, cyclin-dependent kinase-4 and 6, and fibroblast growth factor receptor. Among heterocyclic compounds, pyridopyrimidine compounds are privileged structure in medicinal chemistry, especially with FGFR-TK, CDK-4, ABCG-2, Ef-2 kinase, and Type-II pan-RAF kinase inhibitors (Figure 1). The present review illustrates that pyridopyrimidines have been developed by using new strategies and may be utilized for treatment of various cancer.

Heterocyclic compounds are recognized to possess a high grade of structural diversity and a broad spectrum of therapeutic properties. About two-thirds of the New Chemical Entities approved by the FDA against cancer entails heterocyclic rings and are the foundation stone of medicinal chemistry. Pyrimidine being a major heterocyclic compound and a crucial base component of the genetic material has emerged as the key structural component against cancer, the deadliest disease worldwide. Though lots of drugs are marketed against cancer but researchers still investigating the more promising moieties against various malignancies due to the severity of this disease. In this review, an attempt has been made to assemble the reported literature of the previous five years on various synthetic procedures and anti-cancer potential of various classes of fused pyrimidine analogs which would help the researchers in designing new potent derivatives. Besides this, the review intends to focus on the comprehensive discussion on biological targets, modes of action, and structure-activity relationships of each class of fused pyrimidines as potential anticancer agents.
