The mountain pine beetle (MPB) is an eruptive insect that is currently causing an outbreak of record size in Western Canada. A lack of long distance MPB dispersal data has limited our understanding of and ability to manage MPB epidemics. My goal was to determine the MPBs Western Canadmn population structure. upon wh1ch d1spersal patterns may be supenmposed. I analyzed MPBs from 35 mfested lodgepole pine stands at six microsatellite loc1. The MPB exh1bited strong and s1gmticant Western Canadian population structure. Th1~ population ~tructure wa~ mcongruent w1th the structure of its primary symbiont. 0. £ hn igerwn. but congruent \\1 ith the structure of 1ts primary host, P.contorta. Novel fungal selection pres~ure~ ha"Ve probably caused the discrepancy in beetle/fungus phylogeography. A result of Western Canadmn MPB population structure alternately contrasts and supports population structure~ previou~ly reported for Scolytids, including MPBs. The partitioning of MPB populatiOn structure into a Northern and Southern group is most likely the result of postglacial recolonization and differences in MPB population dynamics. Primarily using my genetic data, I inferred the historical movement patterns of the MPB in Western Canada. I found no evidence that the epidemic spread from an epicenter in Tweedsmuir Provincial Park. My data support multiple sources for the current epidemic; I suggest that regional population expansions have caused the rapid escalation in the severity of the current epidemic. MPB movement patterns and atmospheric wind data were concordant; winds in Western Canada are predominantly westerly or southwesterly. which was the predominant direction of inferred movements amo ng MPB populations. In contrast, current MPB population structure best fits a 30-year climatic suitabi lity distribution for a historical as II opposed to the most current ( 1971 -2000) period. The population genetics of long distance MPB dispersal. an evolutionary theory for MPB population dynamics. and MPB "range expansion" are extensively d1scussed. Potential biases and research limitations are noted. Based on my results and inferences. future areas of investigation are noted.An executive summary. with management recommendations. is prov1ded as a conclusion.

Monitoring arthropod movement is often required to better understand associated population dynamics, dispersal patterns, host plant preferences, and other ecological interactions. Arthropods are usually tracked in nature by tagging them with a unique mark and then recollecting them over time and space to determine their dispersal capabilities. In addition to actual physical tags, such as colored dust or paint, various types of proteins have proven very effective for marking arthropods for ecological research. Proteins can be administered internally and/ or externally. The proteins can then be detected on recaptured arthropods with a protein-specific enzyme-linked immunosorbent assay (ELISA). Here we describe protocols for externally and internally tagging arthropods with protein. Two simple experimental examples are demonstrated: (1) an internal protein mark introduced to an insect by providing a protein-enriched diet and (2) an external protein mark topically applied to an insect using a medical nebulizer. We then relate a step-by-step guide of the sandwich and indirect ELISA methods used to detect protein marks on the insects. In this demonstration, various aspects of the acquisition and detection of protein markers on arthropods for mark-releaserecapture, mark-capture, and self-mark-capture types of research are discussed, along with the various ways that the immunomarking procedure has been adapted to suit a wide variety of research objectives. -The objective of this work was to evaluate the spatial distribution of thrips in different crops, and the correlation between meterological parameters and the flight movements of this pest, using immunomarking. The experiment was conducted in cultivated areas, with tomato (Solanum lycopersicum), potato (Solanum tuberosum), and onion (Allium cepa); and non-cultivated areas, with weedy plants. The areas with tomato (100 days), potato (20 days), and weeds were sprayed with casein, albumin, and soy milk, respectively, to mark adult thrips; however, the areas with onion (50 days) and tomato (10 days) were not sprayed. Thrips were captured with georeferenced blue sticky traps, transferred into tubes, and identified by treatment area with the Elisa test. The dependence between the samples and the capture distance was determined using geostatistics. Meteorlogical parameters were correlated with thrips density in each area. The three protein types used for immunomarking were detected in different proportions in the thrips. There was a correlation between casein-marked thrips and wind speed. The thrips flew a maximum distance of 3.5 km and dispersed from the older (tomato) to the younger crops (potato). As áreas com tomate (100 dias), batata (20 dias) e plantas daninhas foram pulverizadas com caseína, albumina e leite de soja, respectivamente, para marcar os tripes adultos; no entanto, as áreas com cebola (50 dias) e tomate (10 dias) não foram pulverizadas. Os tripes foram capturados com armadilha adesiva de cor azul georeferenciada, acondicionados em tubos e identificados, por área de tratamento, com o teste de Elisa. Determinou-se a dependência entre as amostras e a distância de captura com uso de geoestatística. Os parâmetros meteorológicos foram correlacionados com a densidade de tripes em cada área. Os três tipos de proteína usados na imunomarcação foram detectados, em diferentes proporções, nos tripes. Houve correlação entre os tripes marcados com caseína e a velocidade do vento. Os tripes voaram uma distância máxima de 3,5 km e dispersaram dos cultivos mais velhos (tomate) para os mais jovens (batata).
