An In Vitro Study of the Effect of Cytotoxic Triorganotin Dimethylaminophenylazobenzoate Complexes on Red Blood Cells

Abstract: Interactions of tributyltin (TBTA) and triphenyltin (TPhTA) 2-[4 (dimethylamino)phenylazo]benzoates, showing promising cytostatic activity against tumor cells, with erythrocytes and with erythrocyte membranes and model lipid membranes have been investigated. The effect of TBTA and TPhTA on the erythrocyte and its model membrane was investigated by the microscopic and spectroscopic methods. Interaction of tin complexes with the membrane was determined on the basis of hemolytic activity, changes induced in the s… Show more

“…The tested compounds shifted the maximum of the phosphate band very slightly towards lower values of wave numbers, especially 7b, 8a and 8b for ν s (PO 2 − ) ( Figure 5b) and 6a, 7b, 8a and 8b for ν as (PO 2 − ) ( Figure 5a). Additionally, in the presence of lactones the band of the phosphate group is slightly broader than for pure RBCMs, indicating that there might be an interaction between the investigated compound and the lipid [29]. In addition, preliminary analysis of the amide bands indicates that the tested compounds slightly affect the structure of proteins present in the membrane, which is the subject of our next work.…”

“…The hemolytic activity test was performed using pig red blood cells (RBCs) and their membranes (RBCMs). The method has been described previously by Pruchnik et al [29]. The hemolytic activity of the lactones was tested as DMSO solutions (10,20,40,60,80, and 100 µM concentrations).…”

“…Due to a simple structure and very important functions in the organism, red blood cells (RBCs) are a good model to assess toxicity of different compounds [45,46]. The hemolytic activity test was carried out using RBCs obtained from pig blood, because the percentage content of lipids is closest to the human erythrocyte.…”

“…The method applied was based on that described by Pruchnik et al [45] with minor modifications. The following concentrations of each compound were used: 10, 20, 40, 60, 80, and 100 µM.…”
