Human sex ratio is generally 1.06. The motility selected population of spermatozoa are usually Y chromosome rich. It is therefore easier to influence selected male births. Social and psychologic factors and economic needs intensified by high technology may cause further imbalance in the sex-ratio, creating a demand for techniques to influence sex selection. Such techniques may also help to handle sex linked defects.An examination of several studies on the sex chromosome distribution of human spermatozoa, based on HOP method, showed a preponderance of either X or Y bearing male pronuclei, depending on certain experimental conditions, indicating a technical possibility of influencing sex in an unbiased way. Eine Moglichkeit zur freien Wahl des Geschlechts beim Menschen durch Anreicherung von Xoder Y-Chromosom-tragende SpermatozoenZusammenfassung: Das normale Geschlechtsverhaltnis beim Menschen betragt 1.06. Spermatozoen des Menschen und anderer Sauger, die der Motilitat nach selektiert sind, weisen haufiger ein Y Chromosom auf. Deshalb ist es mit einfachen Methoden moglich, mannliche Nachkommen zu begiinstigen. Psychische und gesellschaftliche Faktoren bevorzugen mannliche Geburten. Wirtschaftliche Bedurfnisse, intensiviert durch die Hochtechnologie, werden das Geschlechtsverhdtnis weiterhin ungiinstig beeinflussen, wodurch der Bedarf an Techniken zur Sex-Selektion enveckt wird.Eine kritische Uberpriifung verschiedener Studien zur Verteilung der Geschlechtschromosomen bei menschlichen Spermatozoen im HOP-Test zeigt eine Haufung von Xoder Y-lialtigen marmlichen Prokernen, was von bestimmten Untersuchungsbedingungen abhangig ist. Dies gibt einen Hinweis auf eine technische Moglichkeit zur freien Wahl des Geschlechts bei der Befruchtung.

Fluorescence in situ hybridization (FISH) with single-color chromosome-specific probes was used to study the rates of disomy for chromosome 1, 16, X, and Y in sperm of fertile and infertile subjects. Diploidy rates were studied using a two-color cocktail of probes for chromosomes 17 and 18 in the same sperm samples. Two-color methodology was not available at the outset of the study. A total of 450,580 spermatozoa were studied from 21 subjects (9 fertile, 12 infertile). Significant differences were observed in the disomy rates between chromosomes with the highest frequency observed for chromosome 16 (0.17%) and the lowest for the Y chromosome (0.10%). No differences were observed between fertile and infertile subjects for either diploidy or disomy. Total disomy rates for chromosomes 1, 16, X and Y ranged from 0.34% to 0.84% among infertile subjects, and 0.32% to 0.61% among fertile subjects. Our data suggest that generalized aneuploidy in sperm is not a major contributor to unexplained infertility.

The mastomys, a small African rodent, has been extensively used as a laboratory animal for cancer research. Previous preliminary studies have demonstrated that the animal has a relatively short estrous cycle and that superovulation can be induced. However, biological characterization of the gametes in this species has not been done. The purpose of this study is to determine requirements for fertilization and to devise a suitable method for in vitro fertilization (IVF). Data from these studies show that 1) cauda epididymal plasma (CEP) is required for maintaining sperm motility in vitro; 2) in a heterologous sperm penetration assay in vitro, optimal conditions for sperm capacitation and the acrosome reaction depend on time and CEP concentration; and 3) loss of the cortical granules is observed to occur spontaneously within 3-4 h after ovulation in superovulated eggs. On the basis of the data obtained from these studies, we have developed a method for preincubation of mastomys sperm and a successful mastomys IVF procedure. This species can now be used as a new laboratory animal for research in reproductive biology.
