The pharyngeal tonsil (adenoid), located at the posterior of the nasopharynx is ideally positioned to sample antigens passing through the nasal cavity or oral cavity. Entering antigens will first contact tonsilar epithelium. To better understand the cellular organization of this important epithelial layer, pharyngeal tonsils were collected from six, 7-month-old calves and examined by light microscopy, immunohistochemistry, and electron microscopy. Morphometric analysis showed that the epithelium overlying lymphoid follicles (reticular epithelium) contained significantly more B-cells, CD4þ, and CD11cþ cells than nonreticular epithelium. In contrast, nonreticular epithelium contained significantly more, c/d TCRþ cells than reticular epithelium. Scanning and transmission electron microscopy of reticular epithelium identified a heterogeneous population of epithelial cells, many of which displayed morphologic characteristics of M-cells. Moreover, putative M-cells were shown to possess the capacity for microparticle uptake. Bovine pharyngeal tonsilar reticular epithelium contains key immune cells, as well as M-cells; elements essential for antigen uptake, antigen processing, and initiation of immune responses. A better understanding of the morphology and function of tonsilar lymphoepithelium will strengthen our understanding of it's role in disease pathogenesis, and potential use as an induction site for mucosal immune responses to vaccination. MALT structure and function in farm animals

-Mucosa-associated lymphoid tissue (MALT) is defined as an organized lymphoid tissue in the mucosa that samples antigens. The morphological characteristics that distinguish MALT from lymphoid infiltrates are discussed. MALT has been extensively investigated in laboratory animals, while knowledge in cattle, sheep, goats, pigs and horses that are summarized under the term farm animals in this review is fragmentary. Literature data about the distribution, morphology, function and involvement in infectious diseases of MALT in farm animals are described. The understanding of specific features of MALT in other species than laboratory animals is important for comparative research, in order to understand pathological and immunological processes in the respective species and as a potential route of vaccination of mucosal surfaces.

Tonsils form a first line of defense against foreign antigens and are also a route of entry and a replication site for some pathogens. The palatine tonsils are the largest of all the tonsils. Despite their general importance, little is known about the microanatomy of the palatine tonsils of the one-humped camel. Palatine tonsils of 10 clinically healthy male camels were obtained directly after slaughtering for human consumption. The tonsils were examined macroscopically and by light, scanning, and transmission electron microscopy. Palatine tonsils had the unique form of several spherical macroscopic nodules protruding into the pharyngeal lumen. These spherical masses were numerous and close together in the lateral oropharyngeal wall, with a few solitary nodules in the dorsal wall. Each nodule had one or two apical openings to crypts, and was enclosed by an incomplete connective tissue capsule and covered apically with stratified squamous keratinized epithelium. The tonsillar crypt was lined with stratified squamous non keratinized epithelium. Several lymphocytes infiltrated the epithelial layer, forming patches of reticular epithelium. Lymphoid follicles with obvious germinal centers extended under the epithelial surface. Diffusely localized lymphocytes were seen in the interfollicular region. High endothelial venules, dendritic cells, macrophages, and plasma cells were observed among these lymphocytes. The unique arrangement of palatine tonsils in separate units with individual crypts results in a very large surface exposed to antigen and indicates a significant immunological role of palatine tonsils in the camel.
