“…Fourth, our current data demonstrate that preanalytical factors known to impact the quality of human tissues 10 , 11 also apply similarly to tissues of nonclinical species like mice (as shown in this article) and presumably other test species as well. The average OD of H&E-stained sections from humans and mice are similar (Supplemental Table 9).…”

“…In a qualitative sense, pathologists routinely make diagnoses across studies for a particular histology method based on its known pattern of staining. However, the authors posit that quantifiable stain attributes of tissue sections slated for DIA may vary substantially over time as reagent lots are replaced and environmental conditions within the facility fluctuate (eg, with the seasons 11 ). Our current results do not address these questions, but our findings do provide an outline of key data categories and QC activities that will be essential considerations in investigating such topics.…”

“…Figure 4 illustrates this analysis. The islet with immunohistochemistry staining was outlined (the 'are of interest,' AOI tool) and the density threshold selection tool was applied to the insulin-positive area estimating their percentage of the islet (Image pro plus version 7.1 for Windows, Media Cybernetics, Rockville, MD, USA) (Chlipala et al, 2019). example we used the Stepanizer web-based application (www.stepanizer.com) to overlap the point system to the pancreas photomicrograph).…”

“…Workflows should be designed to minimize the impact of preanalytical variables on digital pathology and downstream image analysis. 8 The pathologist is uniquely qualified to play an important role in quality control along key steps of the workflow, not only early when it comes to tissue, slides, and staining quality but also in project planning, monitoring ongoing work, and finalizing image analysis projects. While some quality control concepts equally apply to both traditional image analysis and work using AIbased tools, other aspects are unique to each approach.…”
