An evaluation of sequence tagged microsatellite site markers for genetic analysis withinCitrusand related species

Abstract: Microsatellites, also called sequence tagged microsatellite sites (STMSs), have become important markers for genome analysis but are currently little studied in plants. To assess the value of STMSs for analysis within the Citrus plant species, two example STMSs were isolated from an intergeneric cross between rangpur lime (Citrus x limonia Osbeck) and trifoliate orange (Poncirus trifoliata (L.) Raf.). Unique flanking primers were constructed for polymerase chain reaction amplification both within the test cros… Show more

“…Dayanandan et al (1997) showed that SSR loci developed for Pithecellobium elegans were conserved among closely related species, and that there is high potential for the transfer of SSR markers between closed related taxa, as in our study. Conservation of SSRs for closely related plant taxa was also reported by Kijas et al (1995) and Wu & Tanksley (1993). Some reports in animals have also shown similar observations (Moore et al, 1991;Schlotterer et al, 1991;Levine et al, 1995) but sequences were not conserved in humans (Moore et al, 1991) The average size of Shc07 amplification length also seems to correlate well with the molecular phylogeny.…”

“…The allele size ranges were compatible with the data reported in the literature (Kijas et al 1995), with small differences in the absolute size of the base pairs depending on the size of the standard, polymer used in the capillary and/or machine used as shown in the literature (Testolin & Cipriani 2010).…”

“…Printed in Brazil www.sbg.org.br (Viruel and Hormaza, 2004). In Citrus and related genera, microsatellites have been employed for phylogenetic studies (Fang and Roose, 1997;Pang et al, 2003), assessment of genetic variability (Kijas et al, 1995;Corazza-Nunes et al, 2002;Koehler-Santos et al, 2003), identification of zygotic plants (Ruiz et al, 2000;Cristofani et al, 2001;Oliveira et al, 2002) and construction and integration of linkage maps (Kijas et al, 1997;Cristofani et al, 2003) but it is still difficult to identify intraspecific variability in some Citrus species.…”
