An enhanced level of LAMP-2A participates in CD4+T cell hyperactivity in patients with primary biliary cholangitis

Abstract: Background: Primary biliary cholangitis (PBC) is an immune-mediated chronic cholestasis, in which T cell homeostasis plays an important role. Lysosomal-associated membrane protein 2 isoform A (LAMP-2A) has been implicated in the regulation of CD4 + T cell responses. Methods: We comprehensively evaluated the immunobiology of CD4 + T cells in patients with PBC (PBC, n=42), chronic hepatitis B (CHB, n=20), and healthy control subjects (HC, n=20) by flow cytometry including activation status and LAMP-2A expression.… Show more

“…Increased activation of NKT cells, T cells, or NK cells has been reported to contribute to the pathophysiology of cholestasis[ 15 , 19 , 20 ]. Here, we measured and analyzed their percentages and Th1/Th2 cytokine production to evaluate their activation.…”

“…As the most common hepatic disorder, the principal pathogenic factors of EE-induced IHC include estrogen and toxic bile acid, both of which are correlated with immune imbalance. Increased activation status of NKT cells, T cells, or NK cells has been reported to contribute to the pathophysiology of cholestatic diseases[ 15 , 19 , 20 ]. Therefore, the percentages of hepatic NKT cells, CD3 + T cells, and NK cells along with their Th1/Th2 cytokine secretion and bias were compared in the present study.…”

“…Role in PBC. Primary biliary cirrhosis (PBC) is an autoimmune, cholestatic liver disease characterized by 3 main symptoms, i.e., chronic cholestasis, circulating antimitochondrial antibodies (AMA), and distinctive liver biopsy results of nonsuppurative destructive cholangitis and interlobular bile duct destruction [131,132]. Perturbations in gut microbiota can lead to bacterial translocation of its products to the liver due to a leaky intestinal barrier, hence leading to chronic inflammation and fibrosis of the liver.…”
