An Emollient Containing Aquaphilus dolomiae Extract is Effective in the Management of Xerosis and Pruritus: An International, Real-World Study

Abstract: Introduction: Xerosis and pruritus are common manifestations of numerous dermatologic and systemic diseases. We evaluated the effectiveness of an emollient containing an Aquaphilus dolomiae extract (ADE-G1) for the management of pruritus and xerosis in patients of all age with a range of dermatologic and systemic diseases. Methods: This open-label, real-world study involved 5910 patients from 33 European, South American, Asian, and North and South African countries, who applied the product for 7 days twice dai… Show more

“…The active, non-medicated substances are active ingredients that do not qualify as topical drugs [ 66 ], which include saponins, flavonoids, vitamins such as riboflavin and niacin, and beneficial bacterial lysates. Some of these active ingredients were found to improve skin protection, relieve pruritis, exert an anti-inflammatory reaction, exhibit antioxidant properties, and provide biologically essential lipids and antimicrobial activities [ 91 , 92 , 93 ]. Dexpanthenol is approved as a cosmetic ingredient with established safety, and it has evidence-based potent skin-hydration and wound-healing effects [ 19 , 40 , 41 ].…”

“…1,134 An emollient containing Aquaphilus dolomiae (ADE-G1) extract has also been shown to work on scabies itch by inhibiting PAR-2. 135,136 MRGPRX2 (found on non-histaminergic neurons and mast cells) is another potential target of scabies itch. Two novel MRGPRX2 antagonists were found to block substance P-induced de novo synthesis of prostaglandin D 2 and p42/44 mitogenactivated protein kinase signal in human umbilical cord bloodderived mast cells.…”

“…This strain belongs to the family Neisseriaceae within the class Betaproteobacteria, and exhibits specific properties that are beneficial for the skin. A postbiotic, containing a biotechnological extract from this microorganism, was found to display immunomodulatory, anti-inflammatory, antipruritic and tolerogenic activities in pharmacological models of atopic dermatitis, and thus has been incorporated into a skin care emollient [ 13 16 ]. These activities included the modulation of innate immunity by agonizing toll-like receptor (TLR) 2, TLR4 and TLR5, the induction of antimicrobial peptides, the inhibition of cytokine production by T helper (Th) 1, Th2 and Th17 cells, and the inhibition of protease-activated receptor (PAR) 2 and thymic stromal lymphopoietin (TSLP).…”
